Background and Objective: Candidiasis is a kind of disease which is correlated with factorssuch as antibiotics abused, immunodeficiency, pregnancy, estrogen therapy or uncontrolleddiabetes. A rapid and reliable laboratory test involving detection and identification ofCandida species is very important for diagnosis and therapy of candidiasis, but now a moresimple and more reliable technique is shorten for clinic, especially for the vulvovaginalcandidiasis (VVC). Because the components of vaginal mucus are more complicated, detection and identification of Candida as pathogen is also more difficult. In the end of lastcentury when Candida chromogenic medium appeared overseas, which resolves this problemto a certain extent. However, the development of Candida chromogenic medium in ourcountry is absent. Import restricted and costliness usually affect that this medium is used inclinic. To content domestic demands for it and obtain product with higher sensitivity, higherspecificity and higher economy, we are about to develop Candida chromogenic mediumand evaluate it in clinic.Methods: Based on Sabourand glucose agar, an optimal experimental condition is picked outusing enzyme chromogenic substrates (H012, H026, H092) by a series of optimizedexperiments and two teams of orthogonal experiments. It is evaluated whether this mediumhas significant difference from CHROMagar using 166 Candida strains isolated and 212clinical specimens suggestive for VVC. Results.①The components of developed medium: Japanese agar, peptone, malt extract, yeast extract, hemin, glucose, N-acetyl-D-glucosamine, H012, H026, H092, BB2, 2-mercaptoethanol, terramycin, etc.②Evaluated with clinical strains, P>0.05.③Evaluatedwith clinical specimens, P>0.05.Conclusions: There is no significant difference between developed Candida chromogenicmedium and CHROMagar in the hand of capability. It can be used for diagnosis of Candidain clinic.
