Trypan Blue test to detect the death rate of human cholangiocarcinoma cells which are treated with Gemcitabine and 5-Fu.2. To detect the inhibition of human cholangiocarcinoma cells which are treated with Gemcitabine and 5-Fu by MTT assay;3. To observe the cells morphology changes by light microscope and fluorescentmicroscope;4. To detect apoptosis index and cell cycle arrest by flow cytometer.Results: 1. Different concentration of Gemcitabine and 5-Fu can induce apoptosis of cholangiocarcinoma cell line. The apoptosis index depends on time and concentration.2. Different concentration of Gemcitabine and 5-Fu can inhibit proliferation of cholangiocarcinoma cell line.The inhibitation depends on time and concentration. 3. The characteristic apoptosis morphology can be observed, such as cell shrinking,shedding,blowing up,karyopycnosis,chromatic agglutinating nuclear fragmentation and apoptotic body.4. The apoptosis index of cholangiocarcinoma cell line QBC939 gradually increasing with the increase of drug concentration, S stage cells decreases gradually while G1 stage cells increases, G2 stage cells ratio has no obvious change .Conclusions : 1. Gemcitabine and 5-Fu effectively inhibit proliferation of cholangiocarcinoma cell line QBC939 in vitro.2. Gemcitabine and 5-Fu can induce cholangiocarcinoma cell line QBC939 apoptosis invitro.3. Gemcitabine and 5-Fu arrest cell cycle of QBC939 cell line at G1 stage.4.
