Objective To investigate the inhibitory effect of a small.interferingRNA targeting VEGF in nasopharyngeal carcinoma cell line (CNE-2Z) invitro, and explore a new way for the genetherapy of retinal neovascul-arization.Methods Nasopharyngeal carcinoma cells were cultured in vitroand divided into normoxia group (20%) and hypoxia group (5%). AfterVEGF siRNA was transfected into CNE-2Z by Lipofectamine 2000,VEGF expression were tested by immuno-histochemical staining andELISA.Results VEGF 165 and VEGF121 protein expressed in the cytoplasmof CNE-2Z cells under normoxia(brown particle) and the expressionincreased under hypoxia. Under normoxia, the expression of secretingVEGF165 in non-transfection group, pSilencer2.1-U6 hygro transfectiongroup and VEGFsiRNA transfection group shown by ELISA were419.72pg/ml, 410.61pg/ml, 320.76pg/ml respectively; Under hypoxia, theexpression of secreting VEGF165 protein in above three groups were805.61pg/ml, 798.81pg/ml, 496.56pg/ml respectively. VEGFsiRNAdownregulated the expression of VEGF165.Conclusion VEGF specific siRNA can downregulate effectivelythe expression of VEGF protein in CNE-2Z cells in vitro, thus it may be a potential therapeutic approach in retinal Vascularization.
