Objective To set up an efficient, rapid and credible method to detect the expression of CK20 in peripheral blood of patients with colorectal carcinoma and study the relationship between CK20 and micrometastasis of colorectal carcinoma. Analyze mutation of P53 gene in those patients. Method CK20 mRNA in peripheral blood of 36 patients with colorectal carcinomas was detected at various time points before and after operation by fluorescence RT-PCR or nest PCR. Mutation of p53 gene was detected using SSCP. Result The positive rate of CK20 mRNA in peripheral blood before operation was 75%. The positive rate increased to 83.3% in 24 hours after operation. There were 13 positive cases after the short-term chemotherapy, and the positive rate was 36%. No mutation of p53 gene was found out in those patients. Conclusion Fluorescence RT-PCR and nest PCR can detect low expression of CK 20 mRNA in peripheral blood. It is useful to detect micrometastasis of colorectal carcinoma.
