Objective: To study the expression and significance of Survivin andMMP-2 in human renal clear cell carcinoma (RCCC); To investigate therelation between the two genes and angiogenesis in human renal clear cellcarc-inoma (RCCC)Methods: Immunohistochemical method was used to detect Survivin,MMP-2, microvessel density (MVD) in 55 cases of RCCC and 20 cases ofnormal kidney tissues. Age of the patients of RCCC ranged from 29 to 85years.Size of RCCC ranged from 1.7 to 12.3cmResults: (1). The positive rate for expression of Survivin in RCCC was65.5% (36/55), which was higher than that in normal renal tissue (0%, 0/20)(P<0.05). There was relationship between Survivin expression and clinicalstage, tumor grade of differentiation(P<0.05), but no relationship betweenSurvivin expression and tumor size(P>0.05). (2). The positive rate for expr-ession of MMP-2 in RCCC was 61.8% (34/55),which was higher than that innormal renal tissue (25%, 5/20) (P<0.05). There was no relationship betwe-en MMP-2 and tumor size, tumor differentiation grade or clinical stage(P>0.05). (3). There was relationship between Survivin, MMP-2 and lymphon-ode metastasis (P<0.05). (4). there was correlation of the positive expressionbetween Survivin and MMP-2(r=0.767,P<0.05).(5). There was no relation-ship between MVD and tumor size, tumor differentiation grade(P>0.05),There was relationship between MVD and clinical stage or lymphonode metastasi (P<0.05). (6). There was a close relationship between MVD and theexpression of Survivin or MMP-2. MVD in cases with Survivin-positiveRCCC was significantly higher than that in cases with Survivin-negateveRCCC (P<0.05). MVD in cases with MMP-2-positive RCCC was significa-ntly higher than that in cases with MMP-2- negateve RCCC (P<0.05).MVD in cases with survivin and MMP-2-positive RCCC was significantlyhigher than that in cases with Survivin and MMP-2-negative RCCC (P< 0.05).Conclusions:1. It is Survivin gene that may be a useful marker in diagnosis of RCCCand may be an independent predictor of RCCC progression.It may play animpo-rtant role in angiogenesis in RCCC.2. MMP-2 may play important role in occurrence and progression ofRCCC. It may be involved in angiogenesis in RCCC.3. Over-expression of Survivin and MMP-2 indicates poor prognosis. Surv-ivin and MMP-2 can be important diagnostic or therapeutic targets in RCCC...
