Objective:By comparing the expression of tumor necrosis factor -α(TNF-α),tumor necrosis factor receptor 1(TNFR1),and monocyte chemotactic protein-1,MCP-1)in decidua tissue and soluble tumor necrosis factor receptor 1(sTNFR1)in serum between normal pregnancy and spontaneous abortion mouse models,the pathological mechanism of spontaneous abortion was researched..Methods:The abortion-prone CBA×DBA/2 matings were used as the model of spontaneous abortion and nonabortion-prone CBA×BALB/C matings were used as the model of normal pregnancy.The expression of TNF-α,TNFR1 and MCP-1 in decidua tissue of two model groups at day 13 of gestation were detected by immunohistochemistry method.The level of sTNFR1 in serum of two model groups at day 13 of gestation was determined by ELISA.The data was analyzed by t test,χ~2 test and Wilcoxon rank sum test.Results:(1)The rate of embryonic resorption of spontaneous abortion mouse models at day 13 of gestation was significantly higher than that of normal pregnancy mouse models(P<0.05)(2)There was no significant difference in the positive rate of TNF-a between two models.The percentage and intensity of TNF-a protein positive staining of spontaneous abortion mouse models were higher than that of normal pregnancy mouse models.There was significant difference between two models(P<0.05)(3)There was no significant difference in the positive rate of TNFR1 between two models.The percentage and intensity of TNFR1 protein positive staining of spontaneous abortion mouse models were higher than that of normal pregnancy mouse models.There was significant difference between two models(P<0.05)(4)There was no significant difference in the positive rate of MCP-1 between two models.The percentage and intensity of MCP-1 protein positive staining of spontaneous abortion mouse models were higher than that of normal pregnancy mouse models.There was significant difference between two models(P<0.05)(5)The concentration of serum sTNFR1 of spontaneous abortion mouse models was much higher than that of normal pregnancy mouse models.There was significant difference between two models(P<0.05)Conclusion:The expression of TNF-α,TNFR1andMCP-1 in decidua and the quantity of sTNFR1 in serum were associated with spontaneous abortion.The higher expression of TNF-α,TNFR1 and MCP-1 in decidua t might be one of causes of spontaneous abortion.The higher quantity of sTNFR1 in serum was important to maintain gestation.
