| Objective:To discuss the effect of repairing bone defect in rabbits with PRP and particulate of autogenous cancellous bone.Methods:Separating PRP from the 5ml whole blood of rabbits and 0.5ml preservation liquid of blood in tow procedure by centrifugal machine,and count the number of platelet of whole blood and PRP.21 rabbits were divided into 2,4,8 weeks groups at randomly(n=7),in each group,1.5cm bone defect in angle of bilateral mandible were created.One side was randomly chosen as the experimental side,which was filled with platelet-rich plasma combined particulate of autogenous cancellous bone.The other side was only filled with particle of particulate cancellous bone as the control side.After 2,4,8 weeks,investigating of the bone defect was made by means of gross observation,X-ray examination,histology of HE staining, computer graphic analysis in both sides and scanning electronic microscope examination.Under Lane-Sandhu X-ray and histology grade standard,the result of X-ray examination and histology slice investigation were qualified and then analyzed with statistical methods.Result:The number of platelet in PRP is 4.05 times in the whole blood.Two weeks after operation,there are no osseointegration in two sides from radiological and gross examination, and there is no statistical difference between the experiment and control side,but from tissue histology of HE examination,it is different between the two sides(P<0.05).At 4,8 weeks after operation,the rate and quality of new bone formation was statistical different between the experiment and the control side from radiological and tissue histology examination(P<0.01). After 2,4,8 weeks,the area of new bone in experimental side is more than in control side and there is obviously statistical different between the experiment and the control side(P<0.001). From macroscopical study,the defect osseointegration on the experimental side is more than on the control side.Conclusion:PRP can improve the healing of mandibular defect and osteogenesis.It is effective in the repairing of bone defect.PRP and particulate cancellous bone composite may be feasible for bones defect restoration.During this process,the main mechanism of bone healing was bone formation by means of membrance-osteogenisis,bone conduction and bone induction.PRP and particulate cancellous bone composite is synergic in inducing and conducting bone formation.Particulate of autogenous cancellous bone is a good carrier for PRP. |
