Objective Insulin resistant rats induced by lipid were treated with pioglitazone and the effect of pioglitazone on function of vascular endothelial cells were investigated,and elucidated the potential mechanisms involved.Methods 30 male Wistar rats were divided into 3 groups randomly,control group(C,n=10) were fed with commom diet for 12 weeks,IR group(HC,n=10)were fed with high fatty diets for 12 weeks.pioglitazone group.(HC+PIO,n=10)were fed with high fatty diets for 12 weeks and treated with pioglitazone after 6 weeks.fasting blood glucose fasting serum insulin soluble vascular cell adhesion molecule-1 serum nitric oxide were measured respectively,At the end of 12 weeks,the rats were anosthetized,and the thoracic aorta was isolated.EC function was determined bycomparingvasorelaxationtoAch(acetylcholine),an EC-dependent vasodilator,and acidified NaNO2,an EC-independent vasodilator.Results compared with rats fed with a normal diet,rats with high fatty diets for 12weeks,level of fasting blood glucose,fasting serum insulin sVCAM-1 is statistical significance(p<0.05); treatment with pioglitazone in HC group,the level of sVCAM-1 is decreased and there is statistical significance(p<0.05);NO is increased.the rest is not significant difference each other. IR caused asignificant EC dysfunction(maximal relaxation to Ach:50.51%±2.45%vs 99.78%±3.01%C,p<0.01)and improved endothelium dependent vasodilatation.Conclusion The results showed that pioglitazone can improve the endocrine function of endothelia by enhancing.NO activity and increased level of sVCAM-1.
