Condyloma accuminatum is referred to as a venereal or genital wart as iscervical lesions or verruca. Caused by the human papilloma virus, the diseasecan give out no symptoms to be noted by the naked eye. The visible symptomsof the venereal wart may be the following: tumors on the genitals, genitalsores, anal warts, or abnormal vaginal postcoital bleeding, the tipicalbiopsy change is the cytocell presenting in the spinous cells. More than 100different types of HPV exist, most of which are harmless. About 30 types arespread through sexual contact and are classified as either low risk or highrisk. Some types of HPV cause genital warts-single or multiple bumps thatappear in the genital areas of men and women including the vagina, cervix,penis, and rectum. These are considered low risk types. High-risk types ofHPV may cause abnormal Pap smear results and could lead to cancers of thecervix, vulva, vagina, anus, or penis. The recent reaserch has been focusedon the overexpression of some proteins such as HLA—A3B6ODQ2,Caspase—3,Bcl-2,CD44,COX-2. but about overexpression of inhibitor gene ofapoptosis—survivinin is not reported. To detect the expression of a novelinhibitor gene of apoptosis survivinin of condyloma accuminatum and theirrelationshipwithclinicpathological features, we useed immunohistochemicalSASB method in 17 specimens of normal skin tissue,120 of condylomaaccuminatum tissue to study the relation between survivinin and develomentof CA. The research give us some indication: we may seal off survivinin toprevent the unnormal growth of cell to increase curing rate and reduce recurring rate.Objective: we use Polymerase chain reaction (PeR)to determent HPV-DNA oftissue of condyloma accuminatumcollected from anal and genital, the specimenswas maded into biopsy slice, Then detect the expression of a novel inhibitorgene of apoptosis survivinin of condyloma accuminatum by immunohistochemicalSASB method to study the relationship of expression of survivinin withclinicpathological features.Meathod: samples of condyloma aceuminatum were obtained from the patientsin the dermatology department of Tianjin General Hospital from 2005 to 2006.17 specimens of normal skin tissue were obtained from patients in the surgerydepartment with no skin disease. Specimens were in size of 0.2 obtained byscissors, the HPV DNA was determented by Polymerase chain reaction (PCR). allthe specimens was derided into two sections: one is put into formaldehydesolution for 24-48 hours, embedded in paraffin. then made into slice of 4-5um. the other was put into liquid nitrogen(-196℃). expression of survivininin 120 cases of condyloma accuminatum an 17 cases of normal skin tissue biopsyslice is detected by immunohistochemical SASB.Results: PCR: the HPV DNA was determented by Polymerase chain reaction(PCR),the site of the geneal primer amplifition is 6321-6340, the base lenthis 450bp, so the tissue in 450 bp band is positive. Immunohistochemistry:HPVantigenwas assessed by immunohistochemical examination with rabbitanti-bovine papillomavirus, detected by goat antibody against rabbit, thenby rabbit PAP, and stained by DAB. survivinin protein expression was examinedwith a monoclonal antibody, detected by SABC and stained by DAB. 120 casesof Condyloma accuminatum was tested, the positive rate is 52.5%. 17cases ofnormal skin tissue is negative.The difference is marked.Conclusion: 1,HPV might induce Survivin protein over-expression in keratinocytes, mainly located in basal cells and lower and middle spinouscells, and no expression in normal skin. 2,The overexpression of Survivinprotein is positively correlated with the occurrence, the develoment and therecurs of condyloma accuminatum.
