Objective: (1)To observe Endostatin' s direct and synergestic effect with cisplatin on NSCLC cell and illuminate the mechanism. (2) To observe Endostatin' s influence on NSCLC graft' s growth and metastases.Methods: (1) To observe inhibition of xenograft tumor growth, VEGF and MVD in primary tumor in injected site and metastatic tumor in lungs of mice loading high-risk metastatic lung cancer, number of lung metastasis after Endostatin administration and analyze correlation between them. (2) To observe inhibition and cell cycle phase change of Calu-6 cell by Endostatin and DDP and compare their effects.Results:(1) Results of animal experiment showed: strongest inhibition of xenograft tumor growth in cisplatin-treated mice, than Endostatin-treated mice; obvious decrease of VEGF expression (P<0.05) in xenograft tumor and VEGF and MVD in metastatic tumor in lungs (all P<0.05) of mice loading high-risk metastatic lung cancer and number of lung metastasis, positive correlation represented between them. (2) Endostatin can inhibit Calu-6 cells growth with time dependant but not time dependant effect.Conclusion: (1) Endostatin can diminish expression of VEGF in xenograft tumor and both MVD both VEGF and MVD in metastatic tumor, therefore, inhibit distant metastasis. (2) Endostatin can inhibit Calu-6 cells growth with time dependant but nottime dependant effect; single Endostatin can also decrease xenograft tumor in mice, but with effect more slightly than DDP.
