Sea-buckthom is a kind of wild shrub plant. As itself has high medicinal value, it has become the focus of public attention. A large number of studies and achievement by modern science and medical science have made clear that sea-buckthorn were not only the most valuable plant with rich plant in the world up to now, but also had 280 kinds of physiology active substance in its fruit oil and juice, such as organic acid, fatty acid, vitamin etc. Sea-buckthom has the effect of dephlogisticate, disinfect and relieve pain. According to itself special function, people have developed a series of sea-buckthom products. This paper has been done the following work in different sea-buckthorn products with its different composition and content.First, according to the composition and content of organic acid and Vitamin C of sea-buckthorn juice, a method for determination of organic acid and VC of compositions and content in sea-buckthom juice has been completed by RP-HPLC-DAD. The qualitative chemical analysis was performed by identifying of retention time and UV-visible light and the quantitative analysis rely external standard .Nine organic acids (including. oxalic acid, tartaric acid, quinic acid, DL-malic acid, acetic acid, maleic acid, citric acid, fumaric acid, succinic acid) and vitamin C were separated on a ZORBAXSB-C18 column of 30℃with 0.01mol/L (NH4)2HPO4 (PH=2.70) solution as the mobile phase, at the flow-rate of 1.0ml/min. An UV detection at 210nm was used in the determination. Hence we established a determination method for organic acid. The method was accurate with fast determination rate and recovery range was between in 94.2-100.4%. The results showed the main constituent in sea-buckthorn juice were oxalic acid, quinic acid, DL-malic acid, acetic acid.Second, according to the composition and content of fatty acid of sea-buckthom oil, a method for determination of fatty acid of compositions and relative content in sea-buckthorn seed oil and sea-buckthom pulp oil has been completed by GC-MS. The qualitative chemical analysis was performed by identifying of retention time and mass spectrum graph and the quantitative analysis relied area normalization method. The results has showed sea-buckthom seed oil and sea-buckthorn pulp oil had a large number of unsaturated fatty acid, which was beneficial to people, both of them were approximation in composition, but different in content. The results also showed the method was easy to operate with fast determination rate and RSD of precision was smaller than 5% and the qualitative chemical analysis could be quickly performed by identifying of retention time and mass spectrum graph and the quantitative analysis could rely area normalization method.
