Objective:To observe the effects of vitamin B2,vitamin B6 and omega-3 fatty acids and their combination on blood uric acid in rats,and to explore the possible mechanism by studying the pathway of uric acid metabolism.Method:A total of 120 male SD rats were randomly divided into 10 groups,the control group?group C?,the model group?group HU?,the positive control group?group AL?,the vitamin B2 group?group B2,1.18 mg/kg·bw Vit B2 by gavage?,the vitamin B6 group?group B6,23.73 mg/kg·bw VitB6 by gavage?,the VitB2+VitB6 group?group BB,1.18mg/kg·bw VitB2+23.73 mg/kg·bw VitB6 by gavage?,the low-dose omega-3 fatty acids group?group?L,36.75 mg/kg·bw omega-3 fatty acids by gavage?,the middle-dose omega-3 fatty acids group?group?M,73.5 mg/kg·bw omega-3 fatty acids by gavage?,high-dose omega-3 fatty acids group?group?H,147 mg/kg·bw omega-3 fatty acids by gavage?and the VitB2+VitB6+omega fatty acids group?group?B,1.18 mg/kg·bw VitB2+23.73 mg/kg·bw Vit B6+73.5 mg/kg·bw omega-3 fatty acids by gavage?.Rats in the C group were fed with normal feed,and the others were fed with normal feed with yeast and oteracil potassium by gavage.The blood samples were collected to determine the concentration of serum uric acid?SUA?at weeks 0,4 and 12.After the sixteenth week,SUA,serum urea,SCr,activities of adenosine deaminase?ADA?and xanthine oxidase?XOD?in the liver homogenate were detected,and pathological histology of the kidney was observed.The protein levels of organic anion transporters 1?OAT1?and renal-specific transporter?RST?in kidney were detected by western blotting.The combined effects of vitamin B2 and vitamin B6 on hyperuricemia in rats were analyzed based on 2×2 factorial design.Results:After 16 weeks,the result of vitamin supplementation showed that the SUA levels in groups HU,B2,B6 and BB were 369.58 mol/L,281.58 mol/L,290.70 mol/L and294.42 mol/L,respectively;which increased significantly compared with group C?260.50mol/L??p<0.05?.Compared with the HU group,levels of SUA,Urea and SCr in group B2,group B6 and group BB were significantly decreased?p<0.05?;and the activity of XOD in liver homogenate in the AL,the B2,the B6 and the BB groups decreased by16.61%,16.40%,10.18%and 7.39%?p<0.05?,respectively;the activity of ADA in the AL,B2 and B6 decreased by 21.31%,22.51%and 16.19%?p<0.05?,respectively.Comparing with the C group,the level of OAT1 in the HU,B2 and B6 decreased by59.40%,48.12%and 44.36%?p<0.05?,respectively.The RST levels in the HU,AL and BB groups were increased significantly.Comparing with the HU group,the level of OAT1 in the AL group was increased?p<0.05?,and the RST levels in the AL,the B2,the B6 and the BB groups were decreased by 24.17%,55.83%,55.83%and 40.83%?p<0.05?,respectively.There was no indication of interaction between VitB2 and VitB6 by 2×2factorial analysis.The pathological section showed that there were different levels of inflammatory cell infiltration in the HU,the B2,and the BB groups.At the end of the intervention with omega-3 fatty acids,the result showed that levels of SUA in the HU,the?M and the?H groups were increased to 369.58 mol/L,304.50 mol/L and 356.75 mol/L,respectively,being higher than that in group C?260.50mol/L??p<0.05?.Comparing with the HU group,level of SUA in the AL,the?L group and the?M groups were decreased significantly?p<0.05?.There were different levels of inflammatory cell infiltration in the HU and the?H groups.The activities of XOD in liver homogenate in the HU and the?H groups were increased to 78.11U/gprot and75.04U/gprot,which increased significantly compared with group C?p<0.05?.Comparing with the HU group,the activity of XOD in liver homogenate in the AL,the?L and?M groups were significantly decreased?p<0.05?.The activity of ADA was higher in the HU group than that in the C group?p<0.05?.Comparing with the C group,the level of OAT1 in the HU,the?M and the?H groups were decreased by 59.40%,36.84%and51.88%?p<0.05?,respectively;and levels of RST in the HU,the AL,the?L,the?M and the?H groups were increased significantly?p<0.05?.Comparing with the HU group,the level of OAT1 in the AL group was increased?p<0.05?.A combination of vitamin B2,B6 and omega fatty acids showed that the interaction effect on SUA and XOD was antagonism in the mixture intervention at the 12nd and16th week?p<0.05?.Conclusion:Vitamin B2 and B6 can reduce the SUA level in hyperuricemia rats by reducing the generation and reabsorption and promoting the excretion of uric acid.Low-dose and middle-dose omega-3 fatty acids can reduce the SUA level in hyperuricemia rats,while the intervention with high-dose omega-3 fatty acids does not have a beneficial effect on hyperuricemia rats;however the combination of vitamin B2,B6and omega-3 fatty acids cannot reduce the SUA level in hyperuricemia rats.
