Objective: Pinellia ternata has a long history of medicinal utilization in China, Japan, and Korea. Herein, we present a sensitive, selective and robust high-performance liquid chromatography(HPLC) method for simultaneous determination of six organic acids, three nucleosides and ephedrine in P. ternata.Methods: The Rhizoma pinelliae samples were collected from different populations in Southwest China and the ten constituents were detected at wavelength of 210 nm. Chromatographic separation was achieved on a Boston Green ODS reversed-phase column and with a gradient mobile phase of methanol and phosphate buffer(p H 2.3; 10 mmol/l)(v/v) at a fl ow rate of 1.0 ml/min.Results: The range of recovery is between 90.8% and 101.9%, and the calibration curves show high linearity. The relative standard deviations of the new method are less than 3.0% for intra- and inter-day assays. All samples were stable for at least 48 h.Conclusions: A simple, accurate and effective HPLC method was developed for simultaneous determination of six organic acids, three nucleosides and ephedrine in P. ternata. This method was validated to be reproducible and highly sensitive. The new method was successfully applied to qualitative and quantitative analysis of ten samples of P. ternata collected from different places in China. The new HPLC method represents a powerful technique for the quality control of P. ternata.
