Studies of TGF-β1 on the relationship between anti-proliferation and telomerase activity effects on the drug-resistant gastric carcinoma cellsOBJECTIVE: To investigate the effect of TGF-β1 on the anti-proliferation and telomerase activity effects of drug-resistant gastric carcinoma cells.METHODS: Two experimental groups were divided. Control group: SNU601/cis10 cells only. Treatment group: 10ng/mL TGF-β1 was added into the SNU601/cis10 cells, and cultured for 24, 48, and 72h, respectively. The growth of drug-resistant gastric carcinoma cells SNU601/cis10 was observed by reverse-microscopy, the cell inhibition rates of the two groups were detected by MTT at the different time course, and the telomerase activities of two groups were detected by using the TRAP ELISA telomerase detection kit, respectively.RESULTS: MTT method showed that the drug resistant cell survival rate of treatment group was (38.90±2.56)%, (50.00±2.61)% and (50.38±1.86)% at 24, 48, and 72h, respectively, there was correlation between the concentration and time, and significantly increased compared with the control group (p<0.001). The observation by reverse-microscopy showed that the cell proliferation ability was significantly stronger along with the increase of culture time, however, significantlv decreased in the all the different time treatment group. The telomerase activity was positive in control SNU601/cis10 group after 24, 48, and 72h, the△A value is (0.425±0.016), (0.470±0.010) and (0.400±0.022), respectively. The treatment group SNU601/cis10 showed positive for telomerase activity after 24h, but negative after 48 and 72h, the△A value was (0.326±0.008), (0.131±0.006) and (0.123±0.006), respectively. There was significant difference at all different time course between control and experimental groups.CONCLUSIONS: TGF-β1 could inhibit the growth of drug-resistant gastric carcinoma cells, and the inhibition of telomerase activity maybe the one of the mechanisms.
