Uterine cervix cancer is one of malignant tumors of genital system threatening women health and life, which may be resulted from socioeconomic factor, pathological changes of cervix, virus infection and reduction of immune function, and so on. The prevention and cure are difficult because of various pathogenesis. The therapy of uterine cervix cancer mostly uses chemosynthesis drugs at present, but side effects and multidrug resistance of chemosynthesis drugs become frequent in treatment. Naturally occurring drugs have been paid close attention by researcher for their low side-effect and no-drug-resistance. So screening naturally occurring drugs of anti-uterine cervix cancer becomes the study hotspot of pharmacology field.Bidens bipircadta L distributes widespread in China. It is adjuvant of anticancer in common use used by herbalist doctor. The study on pharmacological activity of Bidens bipircadta L had not been developed widely yet. No study on anti-cancer activity of Bidens bipircadta L and its effective constituent was reported.In our study, we investigated anti-uterine cervix cancer activity and its action mechanism of Bidens bipircadta L by animal model of transplanted tumor. The results were as follows. The ethanolic extract from Bidens bipircadta L was prepared by hot refluence method. The effects of ethanolic extract on uterine cervix cancer harboring mice were studied. The results showed that it inhibited the solid tumor growth of solid tumor harboring mice and prolonged the survive of ascites tumor harboring mice. The tumor inhibition rate and the life prolong rate were45.52%,33.97% and 68.5%,46.3% in low and high ethanolic extract groups (21.66 and 43.32 g/kg body weight). It showed that the ethanolic extract from Bidens bipircadta L had anti-uterine cervix cancer action in vivo significantly.The immune organ assay and radioimmunoassay suggested that BBE(21.66 g/kg body weight )significantly increased the thymus and spleen index and BBE(43.32 g/kg body weight) significantly increased the index thymus index. The results showed BBE improved growth of thymus and spleen and increase non-specific immunological function. The BBE is relate to the cervix cancer.The color and texture of liver and kidney in high dose BBE group(43.32 g/kg body weight)were normality and no pathology change observed with eyes. The slices of liver and kidney indicated that cells of liver and kidney disposed compactly and cells structure was distinct. The renal tubules and glomerular was distinct. The results suggested that the BBE was no signs of toxicity to the liver and kidney of tumor harboring mice.With the methods of plant chemical constituent subentry test, we found that Bidens bipircadta L mainly consisted of saccharide, saponin, flavonoids, hydroxybenzene, protein, polypeptide, organic acid, lignin, steride, triterpene, essential oil, quinone, tannin and alkaloid.The flavonoids content was 5.67% in Bidens bipircadta L.Immunohistochemistry showed that BBE up-regulated the expression of bax and p53 protein and down-regulated the expression of bcl-2 and PCNA protein. The results above suggested that apoptosis induced by BBE was modulated by bcl-2 family, p53 and PCNA together.The result of detection by flow cytometry and TEM showed BBE further induced apoptosis of U14 cells.The study in vivo investigated anti-uterine cervix cancer effect of BBE and discussed the action mechanism from the point of view of apoptosis induced by protein expression. It provided rationale to develop Bidens bipircadta L as naturally occurring drug of anti-uterine cervix cancer.
