Objective To investigate the effects of 5,7-dihydroxy- 8-nitrochrysin(8-NOChR) on proliferation and apoptosis in human uterine cervix cancer Hela cell line cultured in vitro .Methods Hela cells were cultured in vitro.MTT assay was used to determine the proliferation of Hela cells.The cell count used to determine the growth of Hela cells. Cell apoptotic percentage were detected by flow cytometry using PI staining. AO/EB fluorescence staining was used to observe the morphologic changes of apoptosis in Hela cell line. DNA agarose gel electrophoresis was used to observe apoptosis of Hela cell line. Expression of intracellular Bcl-2,NF-кB, bax protein was analyzed by Western blotResults MTT assay showed that 8-NOChR significantly suppressed the proliferation of Hela cell line, in a dose-dependant manner and its IC50 was 4.53μM.cell count showed 8-NOChR obviously inhibited growth of Hela cells. flow cytometry using PI staining showed can induction of Apoptosis of Hela cell . Typical morphologic changes of apoptosis could be observed after treatment with 8-NOChR by fluorescence microscope using AO/EB staining. DNA ladder bands appeared in DNA agarose gel electrophoresis after treatment with 8-NOChR at 30μM for 72h. Analysis of intracellular Bcl-2,NF-кB, bax protein showed that 8-NOChR at Various concentration could down-regulate the expression of Bcl-2,NF-кB protein , meanwhile , up- regulate the expression of bax protein.Conclusions 8-NOChR could inhibit proliferation and induce apoptosis in human uterine cervix cancer cells. Its anticancer effect seems to be due to induction of apoptosis ,as a result which down-regulate the expression of Bcl-2,NF-кB protein and up- regulate the expression of bax protein.
