Purpose To investigate the relationship between uPAR.TNF-αand cartilage degradation in experimental osteoarthritis of Rabbits knee .Methods The 48 New Zealand white rabbits were devided into 2 groups as random, 24 experimental, 24 matched control. The experimental animals were injected intraarticularly with uPA in right knee joint, control animals were injected with the same dose fo normal saline. All animals were killed in 4,8,12 weeks respectively after the injections. The rabbit cartilage of the both side of the joints were taken to carry on histology observation(HE) and synovial and blood of experimental osteoarthritis of rabbits knee were taken to carry on Elisa. to detect density of uPAR and TNF-αin the synovial and blood of experimental osteoarthritis of rabbits kneeResults The cartilage destruction was positively correlated with the time. The method was given to value about the histology grade point to each specimen in the light mirror and the organization- histochemication standard(Mankin,1971) was given in the joint cartilage degradation . The result shows comparing both of the experiment animals and the matched controls, there were significant differences between of them. And it does so in the experiment groups. It confirm that experimental osteoarthritis were made successfully.The os uPAR and TNF-αin the synovial and blood of experimental osteoarthritis of rabbits knee was positively correlated with the time., there were significant differences between of them.,P<0.05 And it does so in the experiment groups.P<0.05,and it correlated with the level of cartilage degradation.Conclusion :1.We can copy experimental osteoarthritis of rabbits knee with injection to rabbits knee using uPA.2. TNF-αand uPAR have high density in the synovial and blood of experimental osteoarthritis of rabbits knee. There were significant differences between experimental and matched control..It confirm that the density of TNF-αand uPAR in the synovial and blood were correlated with level of cartilage degradation.3.. TNF-αand uPAR may be correlated in cartilage degradation.,but it is not enough as a index to evaluate cartilage degradation.It is more meaningful to detect unite with other index of cartilage degradation.
