Scutellaria barbata D.Don is perennial herbaceous plant and belongs toScutellari of the family Labiatae. It has been used for medicines for a long timein China. In order to strengthen the comprehensive utilization of ScutellariaBarbata, Scutellaria Barbata polysaccharide's extraction, purification and theactivity of antioxidation were studied. The effect on extraction efficiency wasinvestigated by means of single factor and orthogonal experiments. SBPW andSBPS were gained by using Sephdex and DEAE column chromatography andSBPs's homogeneity was proved by fast protein liquid chromatography (FPLC)and PAGE, its compositon was studied by means of GC-MS. The activity ofantioxidation was studied. The results as follows:1 The parameter of extract technology was perfected; the optimal waterextraction parameters were as follows: tempreture 70℃, extracting 1 times,6hours, solid-liquid ratio 1:30. The yield of Scutellaria Barbata polysaccharidewas 2.703%.2 SBPW and SBPS wre isolated and further purified by DEAE-Cellulose-52and Superdex 200 column chromatography. The IR of them (3420 (O-H),2940(C-H),1665 (C=O),1445 (C-O),1120 (C-O) cm-1) showed characteristicabsorptions of polysaccharide.3 The content of total saccharide of SBPW and SBPS was 89.60%and 92.57%, and the acidic saccharide was 4.9%and 19.6%respectively. SBPs'shomogeneity was proved by fast protein liquid chromatography (FPLC) andPAGE. Its relative molecular weight was estimated to be 1.11×104 by FPLC.GC-MS analysis indicated that it was composed of Rha, Fuc, Ara, Xyl, Man, Glc,and Gal with a molar ratio of 1.00:0.13:0.80:0.10:0.34:0.56:3.04.4 The antioxidant ability of SBPW and SBPS in vitro were studied. Theinhibitions of the SBPs to O2-·and·OH were assayed by the method ofpyrogallol and the Fenton system. SBPW had active ability in scavenging thesuperoxide anion radical and hydroxyl free radical, the IC50 value was 921.73μg/mL and 89.26μg/mL, and SBPs also had active ability in scavenging the superoxide anion radical and hydroxyl free radical, the IC50 value was 800.00μg/mL and 90.42μg/mL respectively. The results indicated that SBPW and SBPShad scavenging effects on hydroxyl radical and superoxide anion radical, hadthe inhibiting effects on lipid peroxidation induced by Fe2+ and H2O2, inhibit thehemolysis of red blood cells. In different antioxidant systems, SBPW and SBPShad different antioxidant ability.5 Aged mice (nine months) were abdominal administrated with SBPW andSBPS at dosages of 100~400mg·kg-1 for 35 days. SBPW and SBPS couldobviously decrease the content of serum and liver lipoperoxide (MDA), and alsoincrease the activity of superoxide dismutase (SOD) of serum and liver in agedmice. The results showed that SBPW and SBPS have an antioxidative effect,suggesting that it has antiageingfunction.
