Studies On The Flavonoid Constituents In Barbata

Scutellaria barbata D.Don is a common-used traditional Chinese medicine.There are a few kinds of substances presenting in Scutellaria barbara D.Don,such as Flavonoids,Alkaloids, Diterpenoids,Steroides,Polysaccharides and so on.The reseach of contemporary pharmacology shows that the coarse extraction,effective parts and effective monomer of Scutellaria barbata D.Don have the pharmacologic activities of antitumor,antibacterial,antimutation, antiinflammation,antifebris,antioxidation and accommodating immunity.This paper reviews the research described so far Scutellaria barbata D.Don profile,including the extraction,sublimation and determination of the Flavonoids compounds,and so on.To study the Flavonoids in Scutellaria barbata D.Don,the thesis includes these parts as follow.To determine the scutellarin and apigenin of Scutellaria barbata D.Don in different districts by LC-MS/MS.The mass spectrometer was operated under the negative ion mode with the ESI source.The transition of m/e(mass charge ratio) 461/285 and m/e(mass charge ratio) 269/117 was used to qualify scutellarin and apigenin,respectively.The linear ranges of calibration curve of scutellarin and apigenin were 2.6-72.7 mg.L^-1 and 2.3-63.8 mg.L^-1,respectively.By using the established LC-MS/MS methods,12 samples of Herba Scutellariae Barbatae in different districts were analyzed.The results showed that the contents of scutellarin and apigenin were 0.03%-0.39 %and 0.02%-0.30%,respectively.To extract and sublime the Flavonoids in Scutellaria barbara D.Don,and then detect by LC-MS. The flavonoids in the Scutellaria barbata D.Don were distilled by ethanol,sublimated by macroporous resin and analyzed by LC-MS.The flavonoids were gotten from the parts desorbing with 20%and 40%ethanol.It was supposed that there were luteolin,5,8,2'-trihydroxy-7-O-glucoside,4'-hydroxywogonin and apigenin in the part desorbing with 40% ethanol.To hydrolyze flavones glycoside into flavones aglycon.The reactant was the part desorbing with water,and the condition of acid hydrolyzation was as follows:water hydrochloride concentration 2 mol.L^-1,temperature 90℃,time 3 h.The production was extracted by n-butanol and separated by MPLC.It was supposed that there were scutellarein,luteolin,apigenin, 4'-hydroxywogonin,and other flavones aglycon in the production.Activity experiment of the anti-tumor activity outside the body to flavones glycoside, fla vones aglycon,the parts desorbing with 20%and 40%ethanol.
