Labeled immnoassay technique can detect body's minim bio-activator by the mensurability of marker and the specificity between antigen/antibody. Along with the application of labelling technique in immuno-study,exact measure of physical chemistry was introduced to immunoassay. Traditional Raman scattering signal is too weak to be a good information readout measure without high sensibility. But surface enhanced Raman scattering (SERS) can amplify the signal by large magnitude. Labeled immunoassay utilizing surface-enhanced Raman scattering has the great selectivity and low detection limit. This new developed labeled immunoassay technique well combines nano-technique,bio-analysis and SERS.This project intends to develop a new technology of immunoassay eventually. A sandwich structure is constructed, in which labeled immuno-coloids and antigen covalently bound to substrate specifically capture different antigens .Through the specific adsorption between antigen and the modified antibody, and Raman spectroscopy effectively enhanced by electromagnetic curbstone between metal substance and colloids, the selective interaction between antigen and antibody can be read out by the labeling molecule. On one hand, modification and regeneration of substrate, selection of marker, detecton limit were researched, we have got much uesful data in these fields. The detection limit was reduced and by using suitable eluting solution, we succeed to accomplish the reusability of captured antigen. On the other hand, we achieve detection from mono-analyte to multi-analyte. By distinguish the characteristic SERS spectrum of the reporter molecules, the presence of analyte is established. Mean while, much parallel experiment have performed.
