[Objective]: To investigate the roles of artesunate on the proliferation and apoptosis of the human colorectal cell line LoVo, also to explore its possible mechanisms of anticancer effects.[Methods]: In vitro, human colorectal cancer cell line LoVo was treated by artesunate and (or) 5-FU in various concentrations, and then the effect of cell growth inhibition was assayed by MTT method; cell cycle and apoptosis was assessed by flow cytometry ; the expression of Survivin,bcl-2 and bax were analyzed by reverse transcriplase polymerase chain reaction and immunocytochemistry(ICC).[Results]: Artesunate could inhibit the growth of the human colorectal carcinoma cell line LoVo, and the inhibitory effect was dose-and-time dependent(P<0.05或P<0.01). There was an apoptosis peak in LoVo cells after incubating with artesunate detected and the ratio of G0/G1 phase decline ,cell mainly arrested in S phase by flow cytometry(P<0.05或P<0.01). Survivin and bcl-2 expressions were downregulated and bax were upregulated significantly (P<0.01).[Conclusion]: Artesunate could inhabit the growth of human colorectal cancer cell line LoVo in a dose-and-time dependent manner, and maybe its mechanisms were arresting cell cycle ; inducing apoptosis through downregulating Survivin and bcl-2 expression and upregulating bax expression .
