Objective : To study the anti-tumor effect and explore its mechanisms of Paeonol(Pae) combined with fluorouracil(5-FU) on the human colorectal carcinoma cell line LoVo.Methods: In vitro,human colorectal cancer cell line LoVo was treated by Paeonol(Pae) and(or)5-FU in various concentrations respectively alone or in combination,and then the effect of cell growth inhibition was assayed by MTT method;the expression of COX-2 (Cyclooxygenase-2),Bcl-2 were analyzed by flow cytometry(FCM).Results: Paeonol(Pae) could inhibit the growth of the human colorectal carcinoma cell line LoVo,and the inhibitory effect was dose-and-time dependent(P<0.05 or P<0.01).Compared with Paeonol(Pae)or 5-FU individual drug group,Paeonol(Pae) combined with 5-FU significantly inhibited the growth of LoVo cells(P<0.01).Typical apaoptotic changes were observed in tumor cells under the light microscope. Compared with Paeonol(Pae)or 5-FU individual drug group,Paeonol(Pae) combined with 5-FU significantly induced apoptosis of LoVo cells(P<0.01).COX-2 and Bcl-2 expression were downregulated;compared with the individual drug groups,in the combine group,the expression of COX-2 and Bcl-2 were downregulated distinctly significantly(P<0.01).Conclusions: Paeonol(Pae) could inhibit the growth of human colorectal cancer cell line LoVo in a dose-and-time dependent manner,Paeonol(Pae) combined with 5-FU could significantly increase the anti-colorectal cancer effect,had synergetic effect, and maybe the anti-tumor mechanisms were inducing apoptosis through downregulating COX-2 and Bcl-2 expression.
