Objective: To study the expression and time course of platelet-derived growth factor-βin cerebral vasospasm after subarachnoid hemorrhage with a rabbit endovascularpuncture model, and to discuss the significance.Material and Methods: Cerebral vasospasm model was produced after SAH byendovascular puncture method. 35 New Zealand white rabbits with health andclean. The SAH groups were divided into seven groups (including 3h,12h,1d,2d,3d,7d,14d), every group has five rabbits. All rabbits were taken vein bloodby each time. The serum levels of PDGF-BB were detected by means of enzyme linkedimmunosorbent assay (ELISA). After perfusion-fixed, sectioned slices were examinedby light microscopy to measure luminal narrowing and wall thickness of posteriorcommunicating artery and basilar artery. The immunohistochemistry technique was used tostudy express of platelet-derived growth factor-βin Smooth muscle cell with basilarartery and posterior communicating artery.Result: Among all, brains of 45 rabbits (including 35 rabbits with subarachnoidhemorrhage) were harvested. Compareing with normal group and nonoperation group,posterior communicating artery and basilar artery showed luminal narrowing and wallthickness. Enhanced expression of PDGF-βprotein at CVS was seen at 3h, and its markedup-regulation was noted at 3-7days, and then the expression became decreased but itremained above the control levels still 14 days after cerebral vasospasm.Conclusion: We demonstrate that this model showes biphasic pattems to theluminal narrowing: acute and delayed constriction phase. The expression of PDGF-β obvious rise and the regular change in time course in cerebral vasospasm aftersubarachnoid hemorrhage. It is important significance in the course of occurrence anddevelopment pathology after cerebral vasospasm.
