OBJECTIVE: To explore whether transplantation of autologous bone marrow mesenchymal stem cells(BM-MSCs) might augment angiogenesis and collateral vessel formation in hindlimb ischemia.METHODS: 12 rabbits which were made left hindlimb ischemia model were devided into 2 groups named experimental group and control group . bone marrow of rabbits were obtained and BM-MSCs were deneshed and cultured in experimental group . When cell population of BM-MSCs reached 10~6 Autologous BM-MSCs were injected into the ischemic skeletal muscles in the experimental group, and DMEM medium for the control group by the same way and in the same part . 4 weeks after cells transplatation,CT angiography and perfusion imaging were performed in the rabbits.RESULTS: (1)All rabbits in control group were found toenail darkening or coloboma;rabbits in experimental group have no serious ischemic appearance.(2)CT angiography shew that collateral arteries were significantly increased in ischemic hindlimbs of experimental group.(3) CT perfusion imaging shew that blood perfusion in ischemic hindlimbs of experimental group was significantly better than control group.CONCLUSION: 1. CTPI which is to take as a new index evaluating collateral vessel formation of hindlimb ischemia model is worth further reseaching.2.We get the preliminary demonstration that Autologous transplantation of BM-MSCs may be a simple and effitive way in the treament of limb ischemia.
