[Objective]: In order to evaluate the protective effects of L-THP to the brain of Status Epileplics of PTZ epilepsy rat models ,we oberved the action ,EEG,and ultrastrcture of acute and chronic PTZ epilepsy rat models which were interfere with L-THP and investigated the opportunity of L-THP could be used to treat epilepsia.[Methods]: 1.Acute epilepsy model: 8 4 healthy SD rats were randomly divided into 6 groups, including the normal group, model group, Diazepam group and the three Rotundine groups at low, middle and high dose respectively. 2. Pentylenetetrazole (PTZ) kindling model: 4 2 healthy SD rats were randomly divided into 3 groups, including the normal group, model group and the middle dose group of Rotundine respectively. The change of epilepsy behavior, electroencephalogram (EEG) and ultrastructure were observed after the intervention of the drugs.[Results]: 1. In acute epilepsy model, the two Rotundine groups at low and middle dose could remarkably extend the latency period of Seizures, decrease attack rate tonic convulsion , cut down the amplitude of electroencephalographic waves when compared with the model group, moreover, the results mentioned above were relevant to the dosage. Contrarily, the high dose Rotundine group could aggravate the Seizure symptoms, decurtate the latency period of Seizures , increase the amplitude of electroencephalographic waves, normal group and Diazepam group all don't induce epilepsy. appearance of ultramicrostructure: the mitochondria were in integrity and the neurons cellular structures were in gear in the normal group. The mitochondria were in illegibility and cristae were broken, with the disruption of the internal and external membrane of the mitochondria, the necrosis or apoptosis neurons cells could be found in the model group. In the two Rotundine groups at low and middle dose, the mitochondria were basically in intact, but the gaps were swelled (esp. the cristae), even some cristae were lost; the neurons cells were changed little. The mitochondria were in cloudy construction, cristae were broken, with the disruption of the internal and external membrane of the mitochondria and the necrosis or apoptosis neurons cells could be found in the high dose Rotundine group. 2.In PTZ kindling model,, the Rotundine group significantly decrease the rate of tonic seizures,and the rat of kindle, decrease the amplitude of electroencephalographic waves of EEG when compared with the model group . appearance of ultramicrostructure: the vacuole degenerations were happened to the majority mitochondria, the amount of mitochondria and neurons cells were both decreased and the necrosis and apoptosis could be found in the model group. Minority mitochondria were swelled, with the vacuole degenerations and the amount of neurons cells was approximately normal in the middle dose Rotundine group.[Conclusion]: 1. the acute experiment and chronic experiment all indicate that L-THP have the effect of anti-epileptic .The actions of Rotundine at certain dosage in the epilepsy rat model induced by Pentyleneterazole are to decrease the level of Epilepsy But the function of Rotundine is weaker than Diazepam's, the actions of high dose Rotundine in the epilepsy rat model induced by Pentyleneterazole are to aggravate the level of Epilepsy. 2. The Rotundine at certain dosage protect hippocampal of the rats which be caused epilepsy by Pentylenetetrazole to avoid damaged. But the high dose Rotundine in the epilepsy rat model induced by Pentylenetetrazole isn't this protection.3. the L-THP at low and middle dose have dose-effect relation.
