Objective: Arachnoid granules or villus is one of the importantorganizational structure formed intracranial meninges, and it played a key rolein the absorption of cerebrospinal fluid and the removal of macromolecularmaterial and particulate. Under the various intracranial pathology, arachnoidgranules or villus may have pathological changes, and resulting its functiondecreased. This study used light microscope and electron microscope toobserve architectonic feature of the arachnoid granules in rat and people, andclear and definite the transifation main way of cerebrospinal fluid, providingbetter theory foundation and basis for morphological for the clinical treatmentof some neurological diseases.Methods:(1)The observation of light microscope about the arachnoidgranules in rats:The left ventricle of rats were perfused with4%formaldehyde,then drow the materials,immersed and fixed, dehydrated, transparented,paraffin imbedding,tissue section, and HE staining, then observed with thelight microscope and collect pictures.(2)The observation of transmissionelectron microscope about the arachnoid granules in rats: The left ventricle ofrats were perfused with2%formaldehyde glutaraldehyde, then drow thematerials, immersed and fixed, repaired the block, prepared ultrathin sections,observed with the transmission electron microscope and taked photos,andaccelerated voltage of80kv.(3)The observation of scanning electronmicroscope about the arachnoid granules about human beings: Cutted thesuperior sagittal sinus of the corpse about specimens, showed arachnoidgranules or villus, immersed and degenerated1week with8%NAOH aqueoussolution, cleaned with distilled water Sufficiently, prepared the samples ofscanning electron microscope,then observed and taked photos, and accelerated voltage of20kv.Results:(1)The results observed by HE staining of the tissue sectionabout the arachnoid granules or villus in rats:Typical arachnoid granulationsor villus charged into the sinus cavity with a finger or spherical. Theattachment of arachnoid granules or villus and transverse sinus is more slender,referred to as the neck, the upper is more wider called the body. The outersurface of arachnoid granules or villus was composed of an epithelial celllayer. Observed carefully under the light microscope, this layer of cells andthe endothelial cells of transverse sinus connectting. The deep surface ofepithelial cell layer surface in arachnoid granules or is fibrous capsule, fibrouscapsule is mainly composed of collagen fibers and elastic fibers. The core ofarachnoid granules or villi is central nucleus, the central nucleus as thenetwork composed of the arachnoid cells and connective tissue fiber, a largenumber of wide hole and lacunae existed within central nucleus. There arevarious cellular around lacuna, consistting of interstitial cells, fibroblasts,macrophages,et al.(2)The results observed by transmission electronmicroscope about the arachnoid granules or villus in rats:In the surface ofarachnoid granules or villus, the epithelial (endothelial) cells is fusiformis,the nucleus of cells slightly humped, the inlaid broad parts of cell and narrowof it is mosaic arrangement. In addition to the structure of nucleus, cytoplasm,cell membrane in the normal cells, in the surface of epithelial (endothelial)cells about arachnoid granules or villus, the cytoplasmic processed into thesinus cavity is visible, the shape of protuberant is microvilli or fine finger,there are branch in the longer protuberant, and it consistent with otherprotuberant. In the cytoplasmic protuberant, there are also a lot of vesicle-likestructures, and it is not of uniform size.There are a large number of pinocytoticvesicles and large vacuoles in the structures of vesicle-like.(3)The resultsobserved by Scanning electron microscope in the arachnoid granules or villusobout human beings: In the superior sagittal sinus, the arachnoid granules orvillus about men appeared with most clusters, the number is Unequal, and themorphology is diversity. The attachment is relatively narrow in arachnoid granules or villus and the wall of superior sagittal sinus, and it was called theneck. In the upper of neck is more enlargement, it was known as the body.After the specimens were dealed with chemically by NaOH, the epitheliumcells (endothelial) were eroded away in the surface of arachnoid granules orvillus, the space truss of connective tissue is clearly visible mainly composedof collagen fibers in the epithelium cells, the support structure of collagenfiber is loose, and a large number of wide hollow and lacuna existed in it.Conclusion:1Within in the venous sinuses of dura mater of rats and human beings(especially the superior sagittal sinus), there are a large number of arachnoidgranules or villus,and Its distribution characteristics is sporadi or cluster; thestructural characteristics of arachnoid granules or villus is covered with a layerof epithelium (or endothelial), and there are a large number of wide hole andthe lacuna in collagen fibers of the subepithelial cells.2In the arachnoid granules or villus, the characteristics of distributionand structural pointed out that it is important installation in backflowing andfiltering of the cerebrospinal fluid, the size of molecules can through, and onlylarger particules and the component of cell were prevented from entering theblood.
