Objective: To detect BRAF protein expression and BRAF V600E mutation in malignant melanoma,and to explore clinical relevance.Methods: EnVision immunohistochemical method was utilized to detect BRAF expression in specimens of malignant melanoma and pigmented nevus.Fluorescence quantitive polymerase chain reaction was utilized to detect BRAF V600E mutation of exon 15 in malignant melanoma.Results: The BRAF expression rate in malignant melanoma was significantly higher than that in pigmented nevus(P<0.05),which was related to age of onset(P<0.05).The frequency of BRAF V600E mutations was 43.33%(13/30)in malignant melanoma,which was also related to age of onset(P<0.05).Conclusion: BRAF high expression and BRAF V600E mutation may play an important role in the prognosis and consequence of malignant melanoma.
