Objectives:To obsearve the effects of the multi-micronutrients on alleviating physical fatigue and improving learning and memorizing ability in KunMing mice aged 4-5 weeks and to explore the mechanisms to provide elemental data for the further study on the school children trial. Methods:Two hundred and twenty KunMing male mice were randomly divided into control group and low, medium, high dosage groups according to their body weight. The dosages were designed 0, 10, 20, 30 times of the recommended intake of multi-micronutrients for school children 2g/ (27kg·d) , i.e. 0, 0.74, 1.48, 2.22g/ (kg·d) . Mice of dosage groups were fed on the hydrous suspension with multi-micronutrients while the mice of control group were fed on the placebo suspension everyday. The body weights of mice were recorded every 3 or 5 days. Each group (control group and low, medium, high dosage groups) was randomly divided into five index groups, include loaded swim test group, Livers' glycogen group, serum urea nitrogen group, serum lactic acid group and step down test group. After step down test, the brains of mice were taken out for determination of neurotransmitter (DA, 5-HT, 5-HIAA, NE) after they were executed.Result:1. Effect of multi-micronutrients on growth in miceIn the growth curve of mice, the body weight gain of mice in each of dosage group was faster than that in control group. Comparing with control group, the body weights of mice in high dosage group were significantly higher from the 19th day on (P < 0.05); the body weights of mice in medium dosage group were significantly higher from the 24th day on (P< 0.01).2. Effect of multi-micronutrients on alleviating physical fatigue in miceIn the experiments to discuss the effect of multi-micronutrients on alleviating physical fatigue, comparing with control group, the glycogen levels of mice in medium dosage group were significantly higher (P < 0.05), and the loaded swimming time of mice in the medium dosage group were significantly longer (P<0.01); the serum urea nitrogen levels of mice in the low dosage group after swimming were significantly lower (P < 0.05).3. Effect of multi-micronutrients on improving the learning and memorizing ability in miceIn the experiment of Step Down Test, comparing with control group, the error times of mice in high dosage group were significantly fewer in memory gain stage (P < 0.05); In memory consolidation stage, the error, times of mice in medium and high dosage groups were significantly fewer (P<0.01; P < 0.05), the latent time of mice in medium dosage group were significantly longer (P < 0.05); In memory fadeaway stage, the error times of mice in low, medium and high dosage groups were significantly fewer (P < 0.05; P<0.01; P < 0.05), the latent time of mice in medium and high dosage groups were significantly longer (P < 0.05; P<0.01).4. Effect of multi-micronutrients on neurotransmitter in brain of miceComparing with control group, the determination of DA in medium group were significantly higher (P < 0.05); the determination of 5-HIAA in each dosage group were higher, especially in low dosage group, the difference has statistics meaning (P < 0.05); the determination of 5-HT and NE in medium and high dosage groups were higher, but the difference has no statistics meaning.Conclusion:1. In experiments of Alleviating Physical Fatigue, comparing with control group, the loaded swimming time of mice in dosage group was significantly longer, and the glycogen levels of mice in dosage group were significantly higher, the serum urea nitrogen levels of mice in dosage group after swimming were significantly lower, we can conclude that the multi-micronutrients play active role in alleviating physical fatigue of mice aged 4-5 weeks. 2. In experiments of Step Down Test, comparing with control group, the error times of mice and rats in dosage group were significantly fewer, the latent time of mice and rats in dosage groups was significantly longer, we can conclude that the multi-micronutrients play active role in improving learning and memorizing of mice aged 4-5 weeks.3 . By the results of neurotransmitter's determination, the effect of multi-micronutrients on improving learning and memorizing in mice because of the increase of DA and 5-HIAA possibly.
