Objective: To investigate the therapeutic effects and its mechanism of tranilaston allergic contact dermatitis, for scientific evidence of treating ACD withtranilast.Methods: Female BALB/c mice were chosen to establish the animal model ofallergic contact dermatitis by the topical application of DNFB. We observedthe inhibition of ear swelling by oral administration of tranilast consecutivelywith different doses, before or after challenge. The levels of IL-2 and IFN-γinmurine serum and the culture supernatant of spleen cell were detected byenzyme-linked immunosorbent assay(ELISA).Results: Murine ear swellings were dose-dependently suppressed, in everydose group of tranilast(P<0.05), no matter given before or after challenge;Comparing with the control, the levels of IL-2 and IFN-γin serum and culturesupernatant of spleen cell were significantly raised(P<0.05). Every dose groupof tranilast could suppress the cytokines in different degrees.Conclusion: As our experiment showed, The levels of IL-2 and IFN-γin themice of ACD were significantly higher than that in the control. It demonstratedthat Th1 type cells played an important part in the mechanism of ACD. Nomatter given before or after challenge, tranilast could dose-dependently inhibitthe ear swelling, and the levels of IL-2 and IFN-γin serum and culture supernatant of spleen cell .The fact that tranilast could inhibit the production ofIL-2 and IFN-γin spleen culture for 24 to 72 hours showed, tranilast tookeffect both in the early and the late stage. In conclusion, tranliast could notonly treat but also prevent the ACD, and the therapeutic effect of tranilast inACD was probably played by inhibiting the expressions of some cytokines.
