Purpose:Study the influence of the shufeng decoction on the Allergic contact dermatitis (ACD) model that is induced by the 2,4-dinitrochlorobenzene(DNCB) by noticing the pathological changes that happen in the pathological tissue, and we can also discuss the immuno-reation that shufeng decoction treat the ACD by measuring the the spleen index thymus index and the level of theγ-interferon (IFN-γ), interleukin-4(IL-4) in the pathological tissue, so that we can provide better reference for the clinical application of the shufeng decoction.Material and method:50 health kunming mice random respectively divided into 5 groups, blank control group, ACD model group, shufeng decoction group, loratadine group, the shufeng decoction with loratadine joint group(i.e. traditional joint group). Making use of the mouse model of the ACD that is induced by the DNCB, then intragastric administration to the laboratory mouse with the shufeng decoction and noticing the changes of the spleen index and thymus index. Using enzyme-linked immunosorbent assay (ELISA) and immuno-histochemical methods to detect the Serum and lesions of IFN-Y, IL-4 levels, from the balance of Th1/Th2 cytokine network to discuss the immunological mechanisms of the Shufeng decoction treating the ACD.Results:1. Shufeng decoction can inhibit the increasing of the spleen index and thymus index in the mouse model of the DNCB-induced ACD.The spleen index and the thymus index of ACD model group was significantly higher than the blank in control group (P<0.01), Shufeng decoction group, loratadine group and the joint group of traditional compare with ACD model group, the thymus and spleen index comparison was significantly reduced (P<0.01 or P<0.05). 2. Shufeng decoction can reduce serum levels of IFN-γ, and increase IL-4 levels in the mouse model of the DNCB-induced ACD.In the ACD model group the serum IFN-y levels were significantly higher than the control group, IL-4 levels were significantly lower than the control group(P<0.01); the serum IFN-γlevels of the Shufeng decoction group, the loratadine group and the combination group of the western medicine and the Chinese medicine are significantly decrease, IL-4 levels were significantly higher(P<0.01).3. Shufeng decoction can reduce the expression of IFN-γand increase expression of IL-4 in the mouse model of the DNCB-induced ACD.The expression of IFN-Y in the ACD model group is significantly higher than the control group, IL-4 expression is significantly lower than control group(P<0.01); in the Shufeng decoction group, the loratadine group and the combination group of the western medicine and the Chinese medicine,the expression of IFN-y significantly is reduced, while the expression of IL-4is significantly ncreased(P<0.01, P<0.05).Conclusion:1. Shufeng decoction can cut down the spleen index and thymus index of the ACD model group induced by the DNCB.2. Shufeng decoction can cut down the content of the IFN-γin the pathological blood serum and tissue.3. Shufeng decoction can increase the expression level of the IL-4 in the pathological blood serum and tissue.
