OBJECTIVES: To study the effect to cardiac function by intramyocardial transplantation of mobilized autologous bone marrow stem cells after myocardial infarction through which created the model of ischemic -myocardial infarction in swines by embolization in the distal end of the left anterior descending branch (LAD) with a gelatin sponge through cardiac catheter, and explore its mechanism initiallyMETHODS: There are 20 healthy Taihu Meishan swines which are divided into four groups (Each one consists of 5 swines) at random to create the model of myocardial infarction. The first group don't give a special processing. The second will be injected G-CSF150 ug/kg.d for continual five days after myocardial infarction. The third one will be injected the G-CSF150 ugs/kg.d for 5 days after 1 week.The fourth one transplant autologous bone marrow mesenchymal stem cells(MSCs) through coronary artery in four weeks after myocardial infarction, and observed the change of each index of cardiac function with MRI, Doppler to each swine before myocardial infarction, transplantation and experimental observative terminus, and detected VEGF and TGF-βnumerus of blood serum in different period after mobilized. It could acquire swine's hearts after eight weeks in the experimental observative terminus, detect the condition of field planting and differentiation in cardiac muscle by the method of immunity histochemistry , and detect the density of blood vessels in cardiac muscle.RESULTS: 1) Compared with A group,left ventricular function, including ejection fraction (EF), left ventricular end-systolic dimension (LVESD), left ventricular end-diastolic dimension (LVEDD), ets. was significantly improved in B group. 2) Serum levels of VEGF and TGF-? in B group were significantly elevated.3) Immunohisto-chemistry study showed that vessels stained withⅧfactor was much more than control group.CONCLUSIONS: 1) The model of myocardial infarction is reliable, reproducible, and similar to the human condition, amenable to investigate other investigation. 2) intramyocardial transplantation of mobilized autologous bone marrow stem cells after myocardial infarction was effective, feasible and safe in ischemic heart disease.3) Bone marrow stem cells were mobilized by G-CSF and migrated to the site of myocardial infarction,and differentiated into viable cardiomyocytes or vascular endothelial..The heart function remarkably was inproved by regeneration of myocytes,inhibition of fibroatrophy and protection of ischemic myocardial structure.
