Objective : To investigate immunohistochemical expression of NY-ESO-1 and MAGE-1,cancer/testis antigens in esophageal squamous cell carcinoma and to correlate their expression patterns with clinicopathologic parameters.To provide some data for tumor-specific immunotheraphy.Methods:Using tissue microarray and immunohistochemical techniques to analyze the expression of NY-ESO-1 and MAGE-1 in 113 cases of esophageal squamous cell carcinoma and 10 normal esophageal tissue.Results:Expression of NY-ESO-1 and MAGE-1 was observed in 32 cases and 41 cases respectively of 109 effective samples of tissue microarray.59(54.1%)expressed at least one CT antigen,13(11.9%)expressed two CT antigens, Immunohistochemical staining was only detectable in the cytoplasm.However the expression of NY-ESO-1 and MAGE-1 in normal esophageal mucosa was negative.NY-ESO-1 and MAGE-1 expression was found more frequently in well differentiated tumors. Statistical significance was found between different pathologic grade(P<0.01).Expression rate of NY-ESO-1 and MAGE-1 has no relationship, however, with tumor size , tumor infiltration depth and lymph node metastasis.Conclusions:NY-ESO-1 and MAGE-1 may serve as target antigens for development of immunotherapy of esophageal squamous cell carcinoma.
