Object: To observe whether early period of the tendon bone healing of rabbit influenced when vascular endothelial growth factor 165 (VEGF165) was injected into the tendon bone tunnel.Methods:The study was performed on twenty healthy, skeletally mature New Zealand White rabbits, The posterior limbs of the rabbit flexor digitorum longus tendon with shin bone on was designed as tendon bone model.These rabbits were randomly divided into two groups.In experiment group,100ng VEGF165 with 0.5 ml PBS was injected into the tendon bone tunnel;In countral group,0.5ml PBS was injected into the tendon bone tunnel.At 2 , 5 weeks after the operation, six specimens in two groups were using biomechanical testing. At the same time,four specimens in two groups were got respectively and the structural integrity of the tendon bone interface were observed using optical microscope.Results :In histological evaluation,the interface between tendon and bone of the experiment group at two week after the operation had relatively compactly integrated , a great quantity of mesenchymals and some newly formed vessels can be found in it,but the countral group of the interface did not heal very much, we can not find any newly formed vessels and newly formed bone cells from the interface. The interface between tendon and bone of the countral group at five week after the operation had very compactly integrated , a great quantity of cartilage cells and some newly formed vessels can be found in it, some skeltogenous cells can be found at the area of bone tunnel.The countral group of the interface had relatively porously integrated, the links between the interface were mainly collagen fibrils,just a little newly formed vessels can be found. In biomechanical testing ,the results of the experiment group and the countral group at two and five week after the operation were be contrasted by matched t-test, the results was significant difference in statistics(P<0.05).Conclusions :VEGF165 can make tendon bone healing to the direction of direct healing,it also can increase the number of newly formed vessels. VEGF165 can promote the early period of tendon bone healing.
