Objective: To study the expression and correlations of maspin,VEGF and MVD in tissue of prostate carcinoma , and discuss their significance in carcinogenesis,progression and angiogenesis of prostate carcinoma, providing a approach for the diagnosis and treatment of the tumor.Methods:: The expression of maspin,VEGF and MVD in 24 normal prostate (NP), 60 benign prostate hyperplasia(BPH) and 58 prostate carcinoma (Pca) was examined with immunohistochemistry .Results: (1) In NP,BPH and Pca,the positive rates of maspin were 87.5%,83.3%,37.9%respectively. The expression of VEGF was on the rise from NP to BPH and Pca, which is 20.8%,53.3% and 72.4% respectively .In NP BPH and Pca,the mean MVD were 16.78±3.68,27.67±2.56,57.95±12.05(P<0.01),it was on the rise from NP BPH and Pca(P<0.01).(2) In Pca group, the expression of maspin is 37.9% , which is relevant to the grading and the clinical staging of prostate carcinoma(P<0.05); the expression of VEGF is 72.4% , which is relevant to the clinical staging of prostate carcinoma (P<0.01),but not to the grading(P>0.05). The mean MVD in Pca group is 57.95±12.05, which is closely related to the grading ,clinical staging and lymph node metastasis of prostate carcinoma(P<0.01).(3) In the Pca group, the expression of maspin and VEGF in the metastasized and non-metastasized lymph nodes groups is 17.4%,51.4%;91.3%,60% respectively . These two proteins is closely related to the lymph node metastasis of prostate carcinoma (P<0.05). MVD in maspin positive and negative group was 47.99±9.60,64.04±1.50 respectively, there was significant difference (P<0.01). (4) The expression of maspin and VEGF is negatively correlated in prostate carcinoma(P<0.01,r=-0.503), maspin and MVD is negatively correlated in prostate carcinoma(P<0.01,r=-0.601), VEGF and MVD is positively correlated in prostate carcinoma(P<0.01,r=0.525).Conclusion: (1) The expression of maspin was significant difference in Pca group, and the expression of maspin is closely related to the grading,the clinical staging and lymph node metastasis in Pca. So the detection of maspin in prostate carcinoma may contribute to the diagnosis and prognosis of prostate carcinoma.(2) VEGF is a phenotype of malignancy in prostate carcinoma, which is closely related to the clinical staging and lymph node metastasis. VEGF is a promoting factor in the development of prostate carcinoma, but can not be an effective index to determine the pathology grading.(3) MVD is closely related to the grading, the clinical staging and lymph node metastasis in Pca. It can be an effective index to determine the pathology grading, the clinical staging and prognosis.(4)VEGF MVD and maspin is negative correlated in prostate carcinoma. A negative correlation was found between the VEGF expression and MVD in primary prostate cancer. Combine VEGF,MVD and maspin can be an effective index of the development and prognosis of prostate carcinoma.
