Objective: To determine the effect of MSCs from rat with different age transplantation on the angiogenesis and heart function after myocardial infarction.Methods: MSCs was collected from bone marrow, cultured and passaged in vitro, then purified by adhesive-screening method. Apoptosis of MSCs were tested under ischemia and anoxia condition .Animals were randomly divided into 3 groups: myocardial infarction control group(n=8),myocardial infarction plus young rat MSCs transplantation group,(n=8) and myocardial infarction plus old rat MSCs transplantation group(n=8).4 weeks later, the different VEGF expression was tested with RT-PCR and Immunohistochemistry, blood vessel density was tested with immunohistochemistry of factor VIII, apoptosis of cardiomyocyte was tested with TUNEL, and heart function of the 3 groups were tested with echocardiography.Results: Under the same ischemia and anoxia condition, there was more apoptotic MSCs from old rat than that from the young . Young rat MSCs transplantation group displayed higher level of VEGF than that of old MSCs transplantation group, and the latter was higher than control group; Young rat MSCs transplantation group displayed more blood vessel than that of old rat MSCs transplantation group, and the latter was more than control group, there was more apoptotic cardiomyocyte in control group than that of old MSCs transplantation group, and the latter was more than young MSCs transplantation group. The heart function of young MSCs transplantation group was better than old MSCs transplantation group, and latter was better than control group.Conclusion: MSCs from young rat can improve heart function better trough angiogenesis and decreasing cardiomyocyte apoptosis than that from the old .
