Objective: To evaluate whether the transplantation of aged and young bonemarrow mesenchymal stem cell in a rat myocardial infarction model could improveheart funotionMethods: Rat bone marrow adherent cultured under sterile conditions to obtainbone marrow mesenchymal stem cells by flow cytometry3rd generation of adherentcells. Cell growth curve, under direct vision will be tested and cells transplantation wasinjected into the lesion model of myocardial infarction.Male SD rats2months of age40,creating a model of myocardial infarction were randomly divided into three groups, theyoung rat bone marrow mesenchymal stem cell transplantation groups, the aged ratbone marrow mesenchymal stem cell transplantation group and blank control group.Press groupings transplant.Transplantation after4weeks detection of cardiac ultrasound(the EF, the FS) by HE staining determination of each group infarct region of the area;detection by immunohistochemical methods BCL-2and of BAX, determinationbetween the groups of myocardial cells and bone marrow mesenchymal stem cellsapoptosis.Results:Adherent cells showed the characteristics of mesenchymal cells:spindle-shaped, swirling arrangement, and presented positive expression forCD90(97.51%);CD44(97.99%);CD29(97.46%);CD31(4.75%);CD34(1.82%);CD45(11.08%).The cell growth curve results show:young rat bone marrow mesenchymal stemcells grow fast than the aged rat bone marrow mesenchymal stem cells. Aftertransplantation4weeks,young rat bone marrow mesenchymal stem cell transplantationgroup, EF, FS significantly improved compared with the preoperative (P <0.05).andsignificantly increased than the old age of rat bone marrow mesenchymal stem cell transplantation group and control group(P <0.05).Transplantation after4weeks,youngrat bone marrow mesenchymal stem cell transplantation in the infarct area, andapoptosis was compared with the elderly rat bone marrow mesenchymal stem cells andthe control group improved significantly.The aged rat bone marrow mesenchymal stemcell transplantation group and control group of cardiac function indexes was nostatistically significant difference (P>0.05).Conclusion:Through the observation of cell morphology and flow cytometryidentification of rat femur bone marrow adherent culture can obtain high purity of bonemarrow mesenchymal stem cells. The comparison of youth and old age in rat bonemarrow mesenchymal stem cell transplantation for the treatment of myocardialinfarction confirmed that bone marrow mesenchymal stem cell transplantation for thetreatment of myocardial infarction, the efficacy decreased with increasing age.
