Objective:In the present study,we examined the expression of FAT10 in colorectal cancer tissue,paracancerous tissue as well as their nomal mucosa.And analysed the relationship between the expression of FAT10 and the character of clinical pathology and the action of biology in colorectal carcinomas.It also have the potential of providing theoretical basis for the prevention,early diagnosis and prognosis of tumor,and have the potential of providing target for the treatment of Gene.Methods:The levels of FAT10 in the tissue were detected by immunohistochemical Elivision plus method and RT-PCR method on protein and mRNA levels in 51 cases of colorectal carcinoma and 51 cases of paracancerous tissue and 51 cases of normal colorectal tissue.Observing the relation between FAT10 expression and the extent of polarization,lymph nodes metastasis,liver metastasis,Duke's clinical stages and diameter of tumor,the position of tumor occurrence.Results:1. By immunohistochemical Elivision plus method,the positive expression rates of FAT10 in colorectal carcinomas were 58.8%, the positive expression rates of FAT10 in paracancerous tissue and normal colorectal tissue were 19.6% and 9.8%.By RT-PCR method, the positive expression rates of FAT10 in colorectal carcinomas were 68.6%, the positive expression rates of FAT10 in paracancerous tissue and normal colorectal tissu were 17.6% and 13.7%.2. Immunohistochemical Elivision plus method and RT-PCR method draw to the same conclusion: The levels of FAT10 in tissue of colorectal carcinomas were obviously higher than that of paracancerous tissue group and normal colorectal group (P<0.05).Compared with the group of no lymph node metastasis, the FAT10 levels of lymph node metastasis group was obviously higher(P<0.05).The intensity of FAT10 expression in tissue of colorectal carcinomas was correlated with liver metastasis(P<0.05). The FAT10 levels were positively related with clinical stages. And the levels of FAT10 had ascending trends according to the clinical stages. The FAT10 levels were no correlated with age, sex, the position of tumor occurrence diameter of tumor, the extent of polarization and other clinicpathological factors.Conclusions:(1).Expression of FAT10 is highly upregulated in colorectal cancer, but not in the paracancerous tissue group and normal colorectal group.(2) The abnormal expression of FAT10 may play important roles in the tumorigenesis and progression of colorectal carcinoma.(3)Expression of FAT10 has the association with lymph nodes metastasis,liver metastasis and the clinic type of pathology.It may have the important significance on forecasting the invasion and metastasis of colon carcinomas.
