Objective:It has been taken more and more attention to the function of the abnormal activation and expression of oncogene for the development of colorectal cancer(CRC).It has been discovered in recent years that there are many kinds of the activation of oncogene such as C-MYC and the inactivation of tumor suppressor gene such as p53 in CRC.Pituitary tumor transforming gene(PTTG)was a newly reported oncogene.Overexpression of PTTG play a great role in promoting cell proliferation,transformation and tumorigenesis,It has been ensured that C-MYC was an important down-stream target for PTTG,and overexpression of C-MYC was related to tumorigenesis including CRC.Here we detected the expression of PTTG in human colorectal cancer and its correlation with C-MYC and its significance in cancer biological characteristics to detect its role in colorectal carnoma.Methods:The expression of PTTGmRNA and PTTG protein were evaluated in nomal colorectal tissures,colorectal adenomas and colorectal cancers by reverse transcription polymerase chain reaction(RT-PCR)and immunohistochemistry(IHC).The result of the exmation was statistically analyzed. Results:1.We has detected the expression of PTTGmRNA in nomal colorectal tissures,colorectal adenomas and colorectal cancers.The expression was significant between these three groups.The expression in colorectal cancers was significantly higher than that in colorectal adenomas and nomal colorectal tissures,and the expression of PTTGm RNA in colorectal adenimas was also significantly higher than in nomal colorectal tissures.2.The expression of PTTG protein in colorectal cancer was significantly higher than in colorectal adenimas and nomal colorectal tissures,and the expression in colorectal adenimas was also significantly higher than in nomal colorectal tissures.We hasn't detected the expression of PTTG protein in nomal colorectal tissures.3.The expression of PTTGmRNA and PTTG protein was related,to Dukes stages.The expression of PTTGmRNA was related to lymph node metastasis.4.There was a positive correlation between the expression of PTTG protein and C-MYC protein in colorectal tissures.Conclusions:PTTG may play an important role in carcinogenesis and development of human colorectal cancer by the investigate of the expression of PTTG in colorectal cancers and its realation with C-MYC.
