Objective:To investigate the expression of PTTG、VEGF、p53、TSP-1。MVD (evaluated by CD34) in Colorectal carcinoma and to explore the relationship beteween PTTG、VEGF、p53、 TSP-1and MVD and clinical parametersMethods:Using immunohistochemical En Vision staining method,the expression of PTTG,VEGF,p53,TSP-1and MVD were detected in18normal tissue,20colorectal adenoma,44colorectal adenocarcinoma, and the statistic method was used to analyze the expression of PTTG correlated with the expression of VEGF,p53, TSP-1and MVD,as well as the relationship of clinnicopatheogical parameters.Results:(1) The frequency of positive expression of PTTG protein in colorectal carcinoma was88.6%and showed higher expression than colorectal adenoma(75%) and normal colorectal tissuel1.1%(p<0.05),and correlated with lymph node metastasis, invasive depth differentiation, intrvenovvascular invasion (p<0.05).(2)VEGF was significantly higher in colorectal carcinoma(70.4%) than in nomal colon tissue and colorectal adenoma. The overexpression level of PTTG in related to lymph node metastasis, invasive depth differentiation, intrvenovvascular invasion (p<0.05).(3)The frequency of positive expression of p53protein in colonrectal carinoma was and much higher than in normal colorectal tissues and colorectal adenoma.Overexression of p53was strongly related to the invsiveness(p<0.05).(4)The expression of TSP-1in normal tissue, colorectal adenoma were significantly higher than that in colorectal cancer(p<0.05),and was only correlated with invasive depth.(5)MVD in colon carcinoma was higher than those in colon adenoma and normal tissue[(67±26vs33±7 and11±4/field, both p<0.05)].(6)There was a postive correlation between expression of PTTG and VEGF, p53,MVD, as well as VEGF,p53, and MVD.There was also negative correlation between expression of PTTG and TSP-1, as well as p53and TSP-1.Conclusions:(1)Overexpression of PTTG,VEGF,p53and low expression of TSP-1maybe play an important role in the development of colonrectal normal (2)The expreesion of PTTG and VEGF were correlated with lymph node metastasis, invasive depth differentiation and vascular invasion.p53and TSP-1were correlated with invasive depth.(3)There were positive correlation between PTTG and VEGF,p53expression in colonrectal cancer.Also there were negtive correlation between PTTQTSP-1,and between p53and TSP-1.We believe that PTTG induce angiogenesis by reducing the expression of TSP-1.Understanding the role may help prevent invasion and metasis of colon cancer.
