Objective To detect the blood Pb,hippocampal volume and height peaks of metabolite NAA,Cho,mI,Glx,Lip and Cr in Pb exposured workers by MRI and MRS,explore the relationship between Pb and early effects of AD.Materials and Methods The exposure group consisted of 15(male 13, female 2)occupational Pb exposed workers from Guangxi smeltery and storage battery factory.The average age was 35.5±7.9(y).The control group consisted of 19 healthy volunteers(male 15,female 2)who never exposed to Pb fume and other intoxicant and the average age was 36.7±9.5(y).The concentration of Pb were measured by graphite furnace atomic absorption spectrometry.All subjects were scanned with PHLIPHS achieve 3.0T superconducting magnetic resonance formatter used for brain MRI and MRS.The experimental results were analysised by SPSS 13.0 statistical software package,and t test,correlation analysis,regression analysisi and chi square test were used.Results The average concentration of Pb fume and Pb dust were respectively 0.43 mg/m~3 and 0.44 mg/m~3 in smeltery,7.14 mg/m~3 and 0.55 mg/m~3 in storage battery factory.All of them were higher than maximum acceptable concentration.Occupational Pb exposed workers had obviously symptom,such as fatigued and weak,ache all over the body and limbs anesthesia.The average level of blood Pb in exposure group was obviously higher than the control group.MRI showed hippocampal volume of left and right in exposure group was obviously lower than the control group(P< 0.01).MRS found that hippocampal NAA/Cr was obviously decreased in exposure group(P<0.05),Lip/Cr was obviously increased(P<0.01).There was inverse correlation between hippocampal volume,NAA/Cr and the level of blood Pb.There was positive correlation between Lip/Cr and the level of blood Pb.Conclusions Hippocampal volume were obviously decreased after Pb exposure,and gradually reduced with the increasing level of blood Pb.The metabolism in hippocamp related with neuron loss or lesion,and cell membrane damaged had been changed after Pb exposure,implying Pb is possibly related with the early effecs of AD. Objective To study the effects of Pb on serumal malondialdehyde (MDA),TTR,the activities of superoxide dismutase(SOD)and glutathione peroxidease(GSH-PX)in occupational Pb exposed workers,in order to explore the relationship between Pb exposure and early effects of Alzheimer disease (AD).Materials and Methods The exposure group consisted of 44 male Pb exposed workers from Guangxi smeltery.The average age was 33.9±7.2(y) The control group consisted of 47 male healthy volunteers who never exposed to Pb fume and other intoxicant.The average age was 33.2±10.2(y).The concentration of Pb was measured by graphite furnace atomic absorption spectrometry.The concentration of MDA,the activeties of SOD and GSH-PX in serum were measured by visible spectrophotometry.The concentration of TTR in serum was measured by immuno-Turbidmetric.Results The average concentration of Pb fume and Pb dust were respectively 0.43 mg/m~3 and 0.44 mg/m~3,which were higher than maximum acceptable concentration.The concentration of blood Pb in exposure group was obviously higher than the control group,and the symptom such as dizziness, fatigued and weak and nausea were obviously higher than control group.SBP had statistic increasing when the Pb concentration of blood was 200ug/L. Correlation coefficient of blood Pb and SBP was 0.303.The levels of serumal MDA in the exposed workers whose blood Pb(BPb)exceed 600ug/L was obviously higher than the control group and the exposed workers whose BPb wewe between 400 and 600ug/L.Yhe activities of serumal SOD in the exposed workers whose BPb between 400 and 600ug/L was obviously higher than the control group.The activities of serumal SOD in the exposed workers whose BPb exceed 600ug/L was obviously lower than the control group and the exposed workers whose BPb wewe between 400 and 600ug/L.The activities of serumal GSH-PX in the exposed workers whose BPb exceed 600ug/L was obviously lower than the control groups and the exposed workers whose BPb wewe between 400 and 600ug/L.The activities of serumal SOD in the exposed workers whose lengths of the exposed standing were between 5 years and 10 years was significantly higher than the control group and the exposed workers whose lengths of the exposed standing were less than 5 years.The activities of serumal SOD in the exposed workers whose lengths of the exposed standing more than 10 years was significantly lower than the exposed workers whose lengths of the exposed standing were between 5 years and 10 years.The effect of Pb on serumal TTR showed no significantly difference.Conclusion Pb may induce the increasing of SBP in workers,also advanced along with the BPb elevates.When BPb is between 400 and 600ug/L, it will increase the activity of serumal SOD.When BPb exceed 600ug/L,the levels of MDA will increase,while the activity of SOD and GSH-PX are obviously decreased.It shows Pb has an obviously toxicity of free radicle.
