Objective: JangTangTongMaiFangObservation of myocardial tissue of diabetic rats pathological changes; exploreeffective JangTangTongMaiFangon experimental diabetic rat heart disease protection.Methods:High-sugar high-fat diet-induced insulin resistance in rats. Then the tail veininjection of a small dose repeatedly streptozotocin(STZ)Establishment of type 2diabetic rat model.Will be a model animals were randomly divided into(3):modelgroup,the Jang Tang Tong Mai Fang (hereinafter:TCM),metformin group(hereinafter:WM group),a separate control group.Medicines Board(13):gavage63mg/ml medicine ingredients of the compound suspension 2ml/kg WM group(12):gavage 27.15mg/ml metformin aqueous solution 2ml/kg model group (12) and thecontrol group were given saline was 2ml/kg, oral treatment once a day, for eightconsecutive weeks. Treatment of rats during eating, drinking water, 100,000 of thegeneral. Eight weeks after the treatment, detection glucose tolerance (IGT) in bodyweight (BW),cardiac index(HW/BW);Detection of total cholesterol(TC), triglycerides(TG),fasting serum insulin (FINS);HE staining using morphological changes ofmyocardial cells,transmission electron microscopy observation myocardial ultra-structure change;Masson staining using myocardial collagen and distribution of crudemyocardial collagen content; using reverse transcriptase-polymerase chain reaction(RT-PCR) myocardial transforming growth factor (TGF-1) mRNA expression.Results:1 A general and biochemical indicators:model rats drink more polyuria moreobvious symptoms of food, glucose, weight, HW/BW compared with the normal control group was significantly(P<0.01), the model proved successful; MedicinesBoard symptoms improved significantly: OGTT, weight and HW/BW decreased(P<0.05) and the effectiveness of western medicine quite(P>0.05) while the levelsof TG, TC, FINS:Compared with model group, Western Group and the ChineseMedicines Board may make FINS, TG (P<0.05, P<0.01) Medicines Board whichreduced TG effective than the medicine group (P<0.01).2 Myocardial tissue morphology changes:optical microscopy, electron microscopyobservation:with the control group, model of myocardial cells myofibrils disarranged,fracture, dissolved; swelling of mitochondria, mitochondrial cristae were disrupted,blank form, muscle glycogen cell lipid droplets can be seen calmly;Chinese medicineand western medicine can be seen myocardial ultrastructure changes to the MedicinesBoard and pathological changes lighter. Masson stain said: rat model of myocardialfibrosis obvious (was L blue),Medicines Board can prevent the formation ofmyocardial fibrosis.3 TGF-β1 mRNA level of myocardial collagen and protein content weredetermined:model group than the control group (P<0.01);Chinese group aboveindicators were significantly lower than those in the model group (P<0.01)Conclusion:The results showed that JangTangTongMaiFang effective components of thediabetic rats with myocardial disease protection.Its mechanism may be:By reducingcholesterol,reduce weight and improve insulin resistance and lower blood glucose,DCM thus corrected the rat heart cells glucose and lipid metabolism disorders;Diabetic rats by inhibiting the myocardial tissue of TGF-β1, inhibition of myocardialextracellular matrix (ECM) accumulation, reduced cardiac index, thereby improvingdiabetic rat heart ultrastructural structure, the prevention of myocardial fibrosis, Earlydiabetic rats inhibited ventricular remodeling. Its lipid-lowering effect than metformingroup hypoglycemic activity and metformin considerable role.
