Objective:To explore the expression of Bad and TβRⅠ,Smad4 in TGF-βpathway and possible mechanisms by which they affect hepatocellular carcinoma. Methods:EnVision immunohistochemistry was used to detect the expression of TβRⅠ, Smad4 and Bad protein in 46 patients with HCC,25 liver Cirrhosis and 19 normal liver tissue.Result:1.The positive rate of TβRⅠprotein was 68.42%(13/19),72.00%(18/25),32.61%(15/46)in normal liver tissue,liver Cirrhosis and hepatocellular carcinoma.The positive rate of TβRⅠwere found to be higher in normal liver tissue 68.42%(13/19)than that in hepatocellular carcinoma 32.61%(15/46),The difference in this two group was significant(P<0.01);The positive rate of TβRⅠwere found to be higher in liver Cirrhosis 72.00%(18/25)than that in hepatocellular carcinoma 32.61%(15/46),The difference in this two group was significant(P<0.01).2.The positive rate of Smad4 protein was73.68%(14/19),68.00%(17/25),30.43%(14/46)in normal liver tissue,liver Cirrhosis and hepatocellular carcinoma.The positive rate of Smad4 were found to be higher in normal liver tissue 73.68%(14/19)than that in hepatocellular carcinoma 30.43%(14/46),The difference in this two group was significant(P<0.01);The positive rate of Smad4 were found to be higher in liver Cirrhosis 68.00%(17/25)than that in hepatocellular carcinoma 30.43%(14/46),The difference in this two group was significant(P<0.01).3.The positive rate of Bad protein was 21.53%(4/19),72.00%(18/25),60.87%(28/46)in normal liver tissue,liver Cirrhosis and hepatocellular carcinoma.The positive rate of Bad were found to be higher in hepatocellular carcinoma 60.87%(28/46)than that in normal liver tissue 21.53%(4/19),The difference in this two group was significant(P<0.01);The positive rate of Bad were found to be higher in liver Cirrhosis 72.00%(18/25)than that in normal liver tissue 21.53%(4/19),The difference in this two group was significant(P<0.01). 4.There is positive correlation between the expression of TβRⅠand Smad4 in hepatocellular carcinoma(P<0.01).Conclusion:The expression of TβRⅠ,Smad4 and bad proteinwas closely related with the occurrence and progress of HCC.
