Object: Study the induced condition of the differentiation from the pancreas ductal epitheliod cells to islet-like cells, the secret function of these cells and the ability of decrease the blood glucose of insulin which secreted by these cells of the normal KM mice by observing the differentiation of the primary culture of the pancreas ductal epithliod cells with glucose-changing medium in vivo.Method: We took the pancreas ductal epitheliod cells of normal KM mice for primary culture and removed the glandular cell by changing the medium consecutively at 24 h,48 h,72 h cause of the different adherence time. Then removed the fibroblast by using medium with 2% FBS and make the pancreas ductal epithelioid cell which can grow in medium free of serum gain dominance. We passage the cell at this condition and induced the third generation cells differentiate by using of the medium without serum. We took the cells which induced for 3 d,5 d,7 d,14 d to identify by immunocytochemistry stain. Using ELISA to detect the ability of insulin-secretion of induced islet-secret cells which grown in high glucose condition for 4 h. Detected the glucose-decrease ability of insulin which secreted by the induced islet-secret cells which grown in high glucose condition for 4 h.Result: The pancreas ductal epithelioid cells induced with medium free of serum for 7 d start differentiating to islet-like cells that positive stain with anti-insulin antibody. The positive cell mult obviously at 14 d following inducing. And these cells can secret insulin in condition of high glucose. The insulin secreted by these cells can decrease the level of glucose in normal KM mice.Conclusion: The pancreas ductal epithelioid cell can differentiate to islet-like cell by induced with medium free of serum. These cells can secret insulin in condition of high glucose. The insulin secreted by these ceils can decrease the level of glucose in normal KM mice.
