Backgrounds Epstein-Barr virus(EBV)is a lymphotropic human gammaherpesvirus that latently infects B lymphocytes, resulting in aconcomitant growth transformation of the infected cell. There areseveral human cancers that closely associated with EBV infection,such as nasopharyngeal, Africa Burkitt's lymphoma, B-cell lymphomaand Hodgkin's disease. Rta is the gene product of BRLF1, which isone of immediate-early gene(IE gene) in EBV lyric cycle. Rta is oneof the most important activator that regulates the switch fromlatency to lyric cycle, its expression can lead to the activationof early genes and ultimately virus replication. Now Rta is theemphasis of the reseach in the world.Objective To detect the expression of protein Rta in gastric carcinoma, esophageal carcinoma, breast carcinoma and lungcarcinoma with specific polyclonal antibodies of the recombinantfusion Rta protein from EB virus, and study the relationship betweenthe lytic EBV and those carcinomas.Methods Rta in 30 cases of gastric carcinoma, 45 cases ofesophageal carcinoma, 42 cases of breast carcinoma and 32 cases oflung carcinoma with adjacent non-neoplastic tissue was detected byimmunohistologic method.Results 6 gastric carcinoma cases, 6 esophageal carcinoma casesand 7 breast carcinoma cases were positive in Rta protein while noRta expression detected in adjacent non-neoplastic tissue. Thedifference between carcinoma and adjacent non-neoplastic tissuewas significant(P=0.012~0.031); 4 lung carcinoma cases werepositive in Rta protein while no Rta expression detected in adjacentnon-neoplastic tissue. The difference between carcinoma andadjacent non-neoplastic tissue was insignificant(P=0.113).Conclusion Rta expression level in gastric carcinoma, esophagealcarcinoma and breast carcinoma is higher than their adjacentnon-neoplastic tissue. The lytic EBV may play a important role inthe pathogenesis of those carcinomas.
