Objectives:1. to investigate the expression of AQP1 of endothelial cell under hypoxiacondition in vitro2. to primarily probe into the possible mechanism of this regulation3. to provide the basis for further studyMethods: The cultured Human umbilical vein endothelial cell stain wererandomly assigned to hypoxia group(the HUVEC was exposed to 1% O2, for3h, 6h, 12h and 24h respectively), and the control group(ordinary culture).AQP1 mRNA expression was detected in HUVEC of all groups by real timePCR and AQP1 protein expression by immunohistochemistry.Results: The AQP1 mRNA and protein expression was significantly increasedin hypoxia group compared with the relevant control group duringhypoxia(from 6h to 12h)(P<0.05). the highest level of AQP1 proteinexpression was in 6h.the level of AQP1 protein expression in hypoxia 3hgroup is 1.4 times of normal group (Z=-2.722, P<0.05);the level of AQP1protein expression in hypoxia 6h group is 2.6 times of normal group(Z=-2.882, P<0.05); the level of AQP1 protein expression in hypoxia 12hgroup is 1.5 times of normal group (Z=-2.562, P<0.05);the level of AQP1 protein expression in hypoxia 24h group has no difference with normal group(Z=-1.6, P>0.05).the level of AQP1mRNA expression in hypoxia 6h groupis 6.9 times of normal group (P<0.05);the level of AQP1mRNA expression inhypoxia 12h group is 3.5 times of normal group (P<0.05);the level ofAQP1mRNA expression in hypoxia 3h and 24h group has no difference withnormal group (P>0.05).Conclusion: Hypoxia can stimulate the expression of AQP1 in vascularendothelial cell. the expression of AQP1 can be seen in the umbilical veinendothelial cell,and the expression of AQP1 protein and mRNA is inceasedafter exposure in hypoxia,then decreased after long exposure, the increase ofAQP1 may be a protective reaction under stress,which may have a role inangiogenesis.
