Objective To determine whether lymphatic target chemotherapy withcarbon nanoparticles absorping 5-FU affects expression of Bcl-2, Bax andCaspase-3 in gastric cancer tissues, metastatic lymph nodes and normalgastric mucosa..Method Twenty-eight patients with gastric carcinoma in our departmentwere divided into lymphatic target chemotherapy group and control groupfrom Octobe 2005 to August 2006. Patients were treated with carbonnanoparticles absorping 5-FU before operation in lymphatic targetchemotherapy group and those were treated with nothing before operation incontrol group. The gastric cancer tissue, metastatic lymph node and normalgastric mucosa were collected after operation. The expression of Bcl-2, Baxand Caspase-3 in those tissues was determined by usingimmunohistochemical technique method.Result The positive expression rates of Bcl-2 in gastric cancer tissues andmetastatic lymph nodes were 28.6% and 25.0%, the positive expressionrates of Bax were 85.7% and 80.0%, the positive expression rates ofCaspase-3 were 57.3% and 55.0% in lymphatic target chemotherapy group;In control group, the positive expression rates of Bcl-2 in gastric cancer tissues and metastatic lymph nodes were 78.6% and 70.0%, the positiveexpression rates of Bax were 28.6% and 30.0%, the positive expression ratesof Caspase-3 were 14.3% and 15.0%. The positive expression rates of Bcl-2in gastric cancer tissues and metastatic lymph nodes in lymphatictargetchemotherapy group significantly lower than those in controlgroup(P<0.05). The positive expression rates of Bax and Caspase-3 ingastric cancer tissues and metastatic lymph nodes in lymphatictargetchemotherapy group were significantly higher than those in controlgroup (P<0.05). In lymphatic target chemotherapy group,the positiveexpression rates of Bcl-2, Bax and Caspase-3 in normal gastric mucosa were14.3%,64.3% and 42.9%,and those were 21.4%,78.6% and 50.0% in controlgroup. The positive expression rates of Bcl-2, Bax and Caspase-3 in normalgastric mucosa were no significantly difference between twogroups(P>0.05).Conclusion The lymphatic targetchemotherapy with carbonnanoparticles absorping 5-FU can down-regulate the expression of Bcl-2 andup-regulate expression of Bax and Caspase-3 in gastric cancer tissues andmetastatic lymph nodes.
