Object To investigate the condition of the interleukin-1 receptor I(IL-1RI), interleukin-1 receptor II(IL-1RII) and interleukin-1 receptor accessory protein(IL-1RAcP) expressed in synovium of osteoarthritis and analyse the biological significance. Methods Immunohistochemistry and reverse transcription-polymerase chain reaction(RT-PCR) were adopted to detect the condition of IL-1RI, IL-1RII and IL-1RAcP expressed in 107 OA patients'synoviums. Results Immunohisto- chemistry shew the strongly positive expression of IL-1RI and IL-1RAcP , and positive expression of IL-1RII . The positive were distributed in lining cells, monocyts and vascular endothelial cells in sublining area, but all of them were negative or weak positive in normal synoviums. RT-PCR shew the expression of IL-1RI, IL-1RII and IL-1RAcP in OA synoviums were significant enhanced then normal synoviums(P<0.05), and the expression of IL-1RI was significant enhanced then IL-1RII(P<0.05), but no significant difference with IL-1RAcP(P>0.05). In II and III degree OA synoviums, the expression of IL-1RI and IL-1RAcP were no significant difference with normal synoviums(P>0.05). The expression of IL-1RII in III degree OA synoviums were significant enhanced then normal(p<0.05). Conclusion IL-1RI, IL-1RII and IL-1RAcP have significant roles in the pathogenesis of OA, especially IL-1RI and IL-1RAcP. But their increase were only observed in the earlier period of OA. These suggest that they maybe have no association with the development of OA and have no direct ratio with severity degree of OA. OA can be cured by interrupting the signal transduction pass by which IL-1 play its biological roles.
