Objective:The aim of this investigation was to make the model rats with acute alcoholic liver damage and the model rats with alcoholic liver disease. Both of the model rats were intervened with Zhihuang decoction, and then measured the level of lipid peroxidation in liver and gastric mucosa, the expression of the ADH1 mRNA, NF-ΚB, COX-2 in liver. To observe the effects of Zhihuang decoction on rats with acute alcoholic liver damage and alcoholic liver disease, and to investigate the possible mechanism.Methods:The experiment was divided into two parts. In the first part Zhihuang decoction was used to treat rats with acute alcoholic liver damage. The levels of SOD and MDA in liver and gastric mucosa were respectively measured by hydroxylamine method and TBA method. Rt-PCR method was used to copy ADH1-mRNA in liver and the concentration was analyzed with software of Bandleader ver3.0. In the second part, Zhihuang decoction was used to treat rats with alcoholic liver disease caused by Liber-Decarli liquid food. The change of rats' liver morphology was observed after being stained by HE, and the expression of NF-κB and COX-2 in liver tissue stained by SP-ELISA were analyzed by semi-quantitative account method. All data were analyzed with SPSS11.5 software. Result:Zhihuang decoction could decline the level of ALT, AST(P<0.01, P<0.01), and could alleviate the pathologic change of liver morphology.The level of SOD in liver and gastric mucosa: in the protective experiment, Zhihuang decoction could activate SOD both in liver and gastric mucosa compared with model group(P<0.01); in the curative experiment, it could activate SOD only in liver (P<0.01).The level of MDA in liver and gastric mucosa: in the protective experiment, Zhihuang decoction could decrease the level of MDA in liver (P<0.05), significantly decrease MDA in gastric mucosa compared with model group (P<0.01); in the curative experiment, it could decrease the level of MDA in liver (P<0.01).The expression of ADH1mRNA in liver: no significant increase was observed in the liver of model rats compared with control group. In the protective group, an obvious increase of the expression of ADH1mRNA was observed compared with model group (P<0.01).The expression of NF-κB in liver: the expression of NF-κB in liver was declined in treating group, compared with that in model group (P<0.01) and control treating group (P<0.05).The expression of COX-2 in liver: the expression of COX-2 in liver had no significant change in treating group, compared with that in model group and control treating group.Conclusion:1. Zhihuang decoction could protect hepatic function and alleviate the morphologic change of liver.2. Zhihuang decoction could dramastically decline the level of lipid peroxidation in the liver of rats with acute alcoholic liver damage, and also decline the lipid peroxidation in gastric mucosa in the protective experiment.3. Zhihuang decoction could greatly enhance the expression of ADH1mRNA in the liver of rats with acute alcoholic liver damage in the protective experiment. 4. Zhihuang decoction could decline the expression of NF-κB in liver of rats with alcoholic liver disease.5. Zhihuang decoction didn't affect the expression of COX-2 in the liver of rats with alcoholic liver disease.
