Background Paraquat is one of the most widely used herbicides in the world.It has been approved for use by authorities in more than 120 countries.Paraquat was discovered in 1955 and introduced to the market place in 1962.Paraquat is a relatively non-selective foliage-applied contact herbicide.It is inactivated on contact with almost all naturally occurring soils and it was this property,perhaps above all others,that provided the greatest breakthrough in chemical weed control at the time of its discovery.The active ingredient paraquat is a non-volatile white crystalline solid, chemical name 1,1-dimethy-4,4-bipyridinium,it is extremely soluble in water and is practically insoluble in most organic solvents.It is highly toxic for humans,and many cases died of acute poisoning.Excessive production of oxygen free radicals has been proposed to play an important role in the pulmonary pathology.But we know little about the pathogenesy of pulmonary fibrosis caused by paraquat.It is imperative for us to understand the reason.Recent study showed that some cytokines may play an important role in the development of acute lung injury.In this paper we try to research the changes of some serum cytokine caused by acute paraquat poisoning.Objective To observe the changes of serum cytokines IL-1β,IL-6,IL-10 and TNF-αin acute paraquat poisoning rats and approach the mechanism of acute lung injury caused by pataquat poisoning.Methods All 72 healthy adult Wistar rats were randomizedly assigned into normal control groups,paraquat high dose grot,p(120 mg/kg),paraquat middle dose(60 mg/kg)group and paraquat low dose group(30 mg/kg).It was divided into three observing segments(8h,24h and 72h)and the standards of serum TNF-α,IL-1β,IL-6 and IL-10 were determined by ELISA assay.The pathological changes were also observed.Results The level of cytokines increased after paraquat poisoning.The cytokines level of poisoning groups was higher than that of control group(P<0.05 or P<0.01)at 24h and 72h.The level of cytokines in paraquat(120 mg/kg)group was higher than that of paraquat(30 mg/kg)group at 72h(P<0.05).All cytokines level of paraquat poisoning groups was higher than that of control group at 8h.The pathology discovered that lung tissue was appear broaden and congestive 8h later and it was more obviously at the 24h later.The appearance of widespread interstitial fibrosis could be found 72h later.Conclusion Paraquat can cause acute lung injury and the influence of cytokines on paraquat-induced acute lung tissue injury maybe poisoning.Xuebijing injection is used widely in treating SIRS caused by other reasons.It could decrease inflammation and inhibit the activity of many cytokines.In this paper we discuss the role of Xuebijing injection in treating paraquat poisoning.Objective To observe the effective of Xuebijing injection in treating acute paraquat poisoning.Methods All 18 healthy adult Wistar rats were randomized assigned into paraquat 250mg/kg poisoning groups,para quat 250mg/kg poisoning + Xuebijing injection (1.72g crude drug/kg)treat groups,paraquat 250mg/kg poisoning + Xuebijing injection(0.86g crude drug /kg)treat groups.Paraquat were given by gastric tube..Xuebijing injection was given through rat tail vein lh later.Arteriovenous blood was drawn 8 h later and lung tissue was remained.Arterial blood gas analysis was determined.Cytokines TNF-α,IL-6,IL-10,IL-1β,PDGF and IGF-1 were determined by ELISA assay and we also observed the pathological changes.Results The PaO2,PaCO2 and pH of Xuebijing injection groups were outstrip than that of non-treat group(P<0.05 or 0.01).There was no difference of TNF-α,IL-6 between treatment and non-treatment groups(P<0.05).Cytokine IL-10,IL-1β,PDG,IGF-1 of treatment group were lower than that of non-treatment group(P<0.05).The pathology discovered that the lung tissue was appear engorge,congestive in non-treatment groups,and many phlegmasia cells were infiltrate in lung tissue.The changes was obviously lighter in treatment groups..Conclusion Xuebijing injection could inhabit cytokines IL-10,IL-1β,PDG and IGF-1 activity and protect the lung injury caused by paraquat.
