Objective To investigate the relationship between the expression of Leptin,PTTG,NCAM and Ki67 in pituitary adenomas and the invasion of pituitary adenomas.Method We immunohistochemically (streptavidin-biotin peroxidase method) detect and analyze the expression of Leptin,PTTG,NCAM and Ki67 in 51 cases with pituitary adenomas(34 cases in invasive group, 17 cases in non-invasive group).Results①The expression of Leptin: The staining of Leptin was located in the cytoplasma of pituitary adenomas cell. The expression of Leptin in 34 invasive pituitary adenomas(IPAs) was 24% and significant lower than that in 17 non-invasive pituitary adenomas(non-IPAs) (P<0.01); In 42 functioning adenomas, the expression of Leptin in invasive group was 22% and significant lower than that in in non-invasive group(P<0.01); In 9 non-functioning adenomas, the expression of Leptin in invasive group was 29% and less significant than that in in non-invasive group(P >0.05).②The expression of PTTG: The staining of PTTG was mainly located in cytoplasma of pituitary adenomas cell. The expression of PTTG in 34 IPAs was 94% and significant higher than that in 17 non-IPAs (P<0.01); In 42 functioning adenomas, the expression of PTTG in invasive group was 96% and significant higher than that in in non-invasive group(P<0.01); In 9 non-functioning adenomas, the expression of PTTG in invasive group was 86% and less significant than that in in non-invasive group(P >0.05).③The expression of NCAM: The staining of NCAM was located in the cytoplasma and membrane of pituitary adenomas cell. The expression of NCAM in 34 IPAs was 85% and less significant than that in 17 non-IPAs (P >0.05); The expression of NCAM in invasive group of functioning adenomas and non- functioning adenomas were respectively less significant than that in in non-invasive group(P>0.05).④The expression of Ki 67: The staining of Ki 67 was located in the nuclei of pituitary adenomas cell. The expression of Ki 67 in 34 IPAs was 85% and significant higher than that in 17 non-IPAs (P<0.01); In 42 functioning adenomas, the expression of Ki 67 in invasive group was also 85% and significant higher than that in in non-invasive group(P<0.01); In 9 non-functioning adenomas, the expression of Ki 67 in invasive group was 86% and less significant than that in in non-invasive group(P>0.05).⑤There was no significant correlation between the expression of Leptin,PTTG and Ki 67 and four parameters(age, sex, tumor size and type). It was negative correlation between the expression of Leptin and the expression of PTTG. in IPAs.Conclusion①The lower expression of Leptin and higher expression of PTTG and Ki 67 in IPAs were significant and Leptin,PTTG and Ki 67 may play a important role in the invasion of pituitary adenomas, especially in the invasive potential of function adenomas, but more investigations in non- function adenomas are necessary. The expression of NCAM was not associated with the invasion of pituitary adenomas, although the expression in IPAs was high.②There was negative coincidence between the expression of Leptin and PTTG in IPAs, it suggest that IPAs be accompanied with modulating obstacle in gene level. The initiating stage in IPAs is oncogene activating and tumor suppressor factor missing ,it result in tumor cell proliferation and invasion occurring. Combinating examination of the expression of Leptin,PTTG and Ki 67 is useful to predict invasiveness,prognosis and recurrence of pituitary adenomas.
